ASAP-MS结合化学计量学鉴别胆类药材及掺伪品OA
Identification of Bile Medicinal Materials and Adulterated Products Based on ASAP-MS Combined with Chemometrics
目的:基于大气压固相分析探头串联单四极杆质谱技术(ASAP-MS),结合化学计量学分析,建立不同来源胆类药材的快速识别以及熊胆粉中掺伪猪胆的辨识模型,为解决胆类药材的真实性鉴定问题寻找新方案.方法:采用ASAP-MS采集熊胆粉、牛胆粉、蛇胆粉、猪胆粉、羊胆粉、兔胆粉、鸡胆粉、鸭胆粉及鹅胆粉的质谱信息,结合化学计量学构建主成分-线性判别分析(PCA-LDA)模型,分别评估不同胆类样品检测结果的可靠性.结果:PCA-LDA模型能够有效区分 9 种胆粉类药材;模型内部验证识别率为 98.35%,外部验证识别率为 97.70%.此外,该方法还可识别熊胆粉中的猪胆粉掺伪,其检识最低限为 3%;模型的内部验证识别率为 99.51%,外部验证识别率为99.22%.结论:该研究建立的ASAP-MS结合PCA-LDA模型能够高效、准确地实现不同胆类药材及猪胆粉掺伪的快速鉴定,为胆汁类药材的真伪鉴定提供了可行的新方法.
Objective:To establish a rapid identification model for bile medicinal materials from different sources and the identifi-cation of Suis Fellis Pulvis in bear bile powder based on atmospheric pressure solid-phase analysis probe tandem single quadrupole mass spectrometry(ASAP-MS)combined with stoichiometric analysis,in order to seek new plans to solve the problem of authenticity identification of bile medicinal materials.Methods:The mass spectrometry information of bear bile powder,cow bile powder,snake bile powder,Suis Fellis Pulvis,sheep bile powder,rabbit bile powder,chicken bile powder,duck bile powder and goose bile powder was col-lected by ASAP-MS.Combined with stoometrics,principal component-linear discriminant analysis(PCA-LDA)model was constructed to separately evaluate the reliability of the detection results of different bile types samples.Results:The PCA-LDA model could effectively distinguish 10 kinds of bile medicinal materials.The internal validation recognition rate of the model was 98.35%,and the external vali-dation recognition rate was 97.70%.In addition,this method could also identify the adulteration of Suis Fellis Pulvis in bear bile pow-der,with a minimum detection limit of 3%.The internal verification recognition rate of the model was 99.51%.The recognition rate of external verification was 99.22%.Conclusion:The ASAP-MS combined with PCA-LDA model established in this study can efficiently and accurately achieve rapid identification of adulteration of different bile medicinal materials and Suis Fellis Pulvis,providing a feasible new method for the authenticity identification of bile medicinal materials.
