三七叶际木霉的分离及其对三七圆斑病的防治研究OA
Isolation of Trichoderma from the Phyllosphere of Panax notoginseng and Application in the Control of Panax notoginseng Round Spot Disease
目的:从三七叶际分离木霉菌,探究其对三七圆斑病的防治效果.方法:采用涂布法初步分离出具有木霉典型特征的真菌,利用两点对峙法评估分离菌株对三七圆斑病病原菌菌丝生长的抑制效果,进一步采用形态学和分子生物学技术鉴定该菌株的属种.利用显微技术研究叶际木霉分生孢子对三七圆斑病菌分生孢子萌发和生长的抑制作用,最后用盆栽法研究叶际木霉对三七圆斑病的防治效果.结果:从三七叶际分离出一株疑似木霉菌,该菌株对槭菌刺孢的抑菌率为 78.33%.根据形态学和ITS序列比对,菌株BY-10 被鉴定为深绿木霉Trichoderma atroviride.马铃薯葡萄糖蛋白胨液体培养基为其分生孢子激活萌发的最适培养基.深绿木霉分生孢子对三七圆斑病菌分生孢子萌发的抑制率为 49.32%.盆栽实验结果表明喷洒深绿木霉分生孢子可以显著降低三七圆斑病的病斑个数.结论:从三七叶际分离了一株深绿木霉,该菌株对三七圆斑病菌的菌丝、分生孢子萌发具有显著抑制作用,可有效减少三七叶上的病斑个数,是一株具有应用潜力的生防菌株,可为三七圆斑病的田间防治提供参考.
Objective:To isolate the Trichoderma from the phyllosphere of Panax notoginseng and explore the control effect in round spot disease of Panax notoginseng.Methods:The fungi exhibiting typical characteristics of Trichoderma were preliminarily isolated using the coating method.The inhibitory effect of the isoloted strain on the mycelial growth of the causal agent of Panax notoginseng leaf spot disease was evaluated using the dual culture method.Furthermore,morphological and molecular biological techniques were em-ployed to identify the genus and species of the strain.Microscopic techniques were used to investigate the inhibitory effects of Trichoder-ma conidia on the germination and growth of conidia of the Panax notoginseng leaf spot pathogen.Finally,a pot experiment was conduc-ted to assess the efficacy of phyllospheric Trichoderma in the prevention and control of Panax notoginseng leaf spot disease.Results:A suspected strain of Trichoderma was isolated from the phyllosphere of Panax notoginseng.This strain exhibited an inhibition rate of 78.33%against Mycocentrospora acerina.Based on morphological characteristics and ITS sequence analysis,strain BY-10 was identified as Trichoderma atroviride.Potato dextrose peptone liquid medium was found to be the optimal medium for the activation and germination of Trichoderma atroviride conidia.The inhibition rate of Trichoderma atroviride conidia on the germination of conidia of the Panax notog-inseng round spot pathogen reached 49.32%.Pot experiments demonstrated that spraying Trichoderma atroviride conidia significantly re-duced the number of lesions caused by round spot disease on Panax notoginseng.Conclusion:A strain of Trichoderma atroviride is isola-ted from the phyllosphere of Panax notoginseng.The strain exhibits significant inhibitory effects on both the mycelial growth and conidial germination of the round spot pathogen,effectively decreasing the number of leaf lesions,it is a biocontrol strain with application poten-tial,the study provides a valuable reference for the field management of round spot disease in Panax notoginseng.
