目的:针对土鳖虫药材中雌雄地鳖个体混用的现象,建立一种快速、准确的地鳖性别分子鉴定方法,为其药材质量控制提供技术支持.方法:利用Illumina高通量测序技术,分别测定雌、雄地鳖成虫线粒体基因组全序列;比对分析雌、雄个体间的线粒体基因组差异位点,并进行测序验证及酶切位点分析;基于特异酶切位点,建立PCR-RFLP分子鉴定方法.结果:成功获得雌、雄地鳖各 3 条线粒体基因组序列,全长均为 15 551 bp,雌、雄个体间存在 3 个稳定的碱基差异位点(第 4 113、4 413、8 262 位点).基于差异位点所在片段设计的两对通用引物能够有效扩增目标序列,进一步利用EcoT14 I酶切位点成功建立了PCR-RFLP鉴定体系,该方法可在2.5 h内完成地鳖样品的性别鉴定.结论:该研究所建立的PCR-RFLP性别鉴定方法,准确度高,方便、快捷,为土鳖虫药材性别的质量控制与标准的制定提供了可行的技术方案.

Objective:To establish a rapid and accurate molecular gender identification method for the mixed use of female and male Eudolyphaca sinensis individuals in the medicinal materials of Eupolyphaga Steleophaga,providing a technical support for the qual-ity control of the medicinal materials.Methods:The complete mitochondrial genome sequences of adult female and male Eupolyphaga sinensis were determined respectively by using Illumina high-throughput sequencing technology.The mitochondrial genomic difference loci between female and male individuals were compared and analyzed,and sequencing verification and enzyme digestion site analysis were conducted.Based on specific enzyme digestion sites,a PCR-RFLP molecular identification method was established.Results:Three mitochondrial genome sequences of female and male Eupolyphaga sinensis were successfully obtained,with a total length of 15 551 bp.There were three stable base difference sites(sites 4 113,4 413,8 262)between female and male individuals.Two pairs of universal primers designed based on the fragment where the difference site was located could effectively amplify the target sequence.Further,the PCR-RFLP identification system was successfully established by using the EcoT14 I enzyme digestion site.This method could complete the gender identification of Eupolyphaga sinensis samples within 2.5 hours.Conclusion:The PCR-RFLP gender identification method established in this study is highly accurate,convenient and fast,providing feasible technical solution for the quality control and standard formulation of the gender of Eupolyphaga Steleophaga medicinal materials.
