调脾护心方通过AKT/mTOR通路促进自噬抑制巨噬细胞泡沫化及焦亡的作用研究OA

目的:探讨调脾护心方对氧化低密度脂蛋白(ox-LDL)诱导的巨噬细胞泡沫化和焦亡的作用及机制.方法:体外培养人单核-巨噬细胞系THP-1,使用ox-LDL刺激诱导使其成为泡沫细胞,并通过噻唑蓝(MTT)比色法确定调脾护心方对巨噬细胞适当给药浓度后,使用不同浓度调脾护心方进行干预.油红O染色观察细胞内脂质沉积,酶法测定细胞内胆固醇水平,Western Blot检测巨噬细胞 ABCA1、ABCG1、NLRP3、GSDMD、IL-1β、LC3、P62、p-AKT、p-mTOR蛋白表达,自噬双标腺病毒检测自噬流.利用自噬抑制剂 3-MA抑制细胞自噬,观察调脾护心方是否通过调控自噬抑制ox-LDL诱导的巨噬细胞泡沫化和焦亡.结果:调脾护心方(10~100 mg/L)对巨噬细胞无毒性.与模型组比较,调脾护心方能减少巨噬细胞中脂质沉积和TC、CE的含量,上调ABCA1、ABCG1 蛋白表达,下调NL-RP3、Cleaved GSDMD、Mature IL-1β、P62、p-AKT、p-mTOR蛋白表达,升高LC3 Ⅱ/LC3 Ⅰ比值,并能明显增强自噬流(P<0.05 或P<0.01).3-MA可逆转调脾护心方的作用(P<0.05 或P<0.01).结论:调脾护心方可能通过抑制AKT/mTOR信号通路来提高巨噬细胞自噬活性,从而促进胆固醇外流来减少泡沫细胞的形成,并抑制ox-LDL诱导的巨噬细胞焦亡.

Objective:To investigate the effect and mechanism of Tiaopi Huxin Decoction on oxidized low-density lipoprotein(ox-LDL)-induced macrophages foaming and pyroptosis.Methods:The human mononuclear-macrophage cell line THP-1 was cultured in vitro.It was stimulated and induced to become foam cells by ox-LDL.The appropriate administration concentration of the Tiaopi Huxin Decoction for macrophages was determined by the MTT colorimetric method.Different concentrations of the Tiaopi Huxin Decoction were used for intervention.Intracellular lipid deposition was observed by oil red O staining.Intracellular cholesterol levels were determined by enzymatic method.The protein expressions of ABCA1,ABCG1,NLRP3,GSDMD,IL-1β,LC3,P62,p-AKT and p-mTOR in macrophages were detected by Western Blot.Autophagy flow was detected by double-labeled adenovirus.Autophagy was inhibited by the autophagy inhibitor 3-MA to observe whether the Tiaopi Huxin Decoction inhibited the foaming and pyroptosis of macrophages induced by ox-LDL by regulating autophagy.Results:Tiaopi Huxin Decoction(10 to 100 mg/L)was non-toxic to macrophages.Compared with the model group,Tiaopi Huxin Decoction could reduce lipid deposition and the contents of TC and CE in macrophages,up-regulate the protein ex-pressions of ABCA1 and ABCG1,and down-regulate the protein expressions of NLRP3,Cleaved GSDMD,Mature IL-1β,P62,p-AKT,and p-mTOR.The ratio of LC3 Ⅱ/LC3 Ⅰ was increased,and autophagic flux was significantly enhanced(P<0.05 or P<0.01).The effect of the 3-MA could reverse the effect of Tiaopi Huxin Decoction(P<0.05 or P<0.01).Conclusion:Tiaopi Huxin Decoction may enhance the autophagic activity of macrophages by inhibiting the AKT/mTOR signaling pathway,thereby promoting cholesterol effervesia to reduce the formation of foam cells and inhibiting ox-LDL-induced pyroptosis of macrophages.
