白虎汤对蛋白泛素化与抗原递呈的影响OA
Effect of Baihu Tang on Protein Ubiquitination and Antigen Presentation
[目的]通过研究白虎汤对炎性细胞中焦亡相关蛋白全长gasdermin D(full length gasdermin D,GSDMD-FL)和 Pro-caspase-1 的泛素化及对 2 种蛋白与主要组织相容性复合体Ⅱ(major histocompatibility complex,MHC Ⅱ)分子相互作用的影响,探究经典方剂白虎汤对内源性抗原递呈过程的调节作用.[方法]分别在小鼠肺泡巨噬细胞(MHS)和骨髓源巨噬细胞(BMDM)中建立焦亡细胞模型,并以白虎汤抑制细胞焦亡,利用免疫共沉淀技术检测细胞中内源性抗原GSDMD-FL和Pro-caspase-1的泛素化变化;通过Western blotting检测白虎汤干预前后细胞中MHC Ⅰ和MHC Ⅱ蛋白表达;通过体外蛋白互作研究,分析白虎汤干预对MHC Ⅱ与GSDMD-FL和Pro-caspase-1互作的影响.[结果]在MHS和BMDM细胞焦亡模型中,0.75~3 mg/mL白虎汤均显著抑制细胞中乳酸脱氢酶(lactate dehydrogenase,LDH)和白介素-1β(interleukin-1β,IL-1β)的释放(P<0.05),并促进细胞焦亡相关蛋白GSDMD-FL和Pro-caspase-1的泛素化水平(P<0.05).同时,白虎汤也显著促进了焦亡细胞中MHC Ⅱ的表达,抑制了MHC Ⅰ的表达(P<0.05),并具有一定的剂量效应.体外蛋白互作结果显示,白虎汤显著增强MHC Ⅱ蛋白与内源性抗原GSDMD-FL或Pro-caspase-1的相互作用,促进抗原递呈.[结论]白虎汤促进内源性抗原的泛素化-MHC Ⅱ递呈途径是其清除细胞焦亡危险因素的重要作用机制.
[Objective]By studying the ubiquitination of full length gasdermin D(GSDMD-FL)and Pro-caspase-1 in inflammatory cells and the interaction between the two proteins and major histocompatibility complex Ⅱ(MHC Ⅱ)molecules,the regulatory effect of the classic formula Baihu Tang on the endogenous antigen presentation process was investigated.[Method]Pyroptotic cell models were established in mouse alveolar macrophages(MHS)and myeloid-derived macrophages(BMDM),respectively.Pyroptosis was inhibited by Baihu Tang.The ubiquitination changes of endogenous antigens GSDMD-FL and Pro-caspase-1 in the cells were detected by co-immunoprecipitation technology.The expression of MHC Ⅰ and MHC Ⅱ proteins in cells before and after the intervention of Baihu Tang were detected by Western blotting.Through in vitro protein interaction studies,the effect of Baihu Tang intervention on the interactions between MHC Ⅱ and GSDMD-FL and Pro-caspase-1 was analyzed.[Result]In the pyroptosis models of MHS and BMDM,0.75-3 mg/mL of Baihu Tang significantly inhibited the release of lactate dehydrogenase(LDH)and interleukin-1β(IL-1β)in cells(P<0.05),and promoted the ubiquitination levels of pyroptosis-related proteins GSDMD-FL and Pro-caspase-1(P<0.05).Meanwhile,Baihu Tang also significantly promoted the expression of MHC Ⅱ in pyroptotic cells and inhibited the expression of MHC Ⅰ(P<0.05),with a dose-dependent manner.In vitro protein interaction results showed that Baihu Tang significantly enhanced the interaction between MHC Ⅱ and endogenous antigens GSDMD-FL or Pro-caspase-1,promoting antigen presentation.[Conclusion]The mechanism by which Baihu Tang promoted the ubiquitination-MHC Ⅱ presentation pathway of endogenous antigens was an important role in eliminating the risk factors of pyroptosis.
