[目的]了解甘肃地区牛群中牛病毒性腹泻病毒(Bovine viral diarrhea virus,BVDV)的流行株情况及其分子特征,为该地区BVDV防控提供参考.[方法]对前期检测为BVDV阳性的血清样本进行病毒分离,并利用靶向BVDV NS3蛋白的单克隆抗体对分离毒株进行间接免疫荧光试验(IFA)鉴定.根据分离株全基因组测序结果,对其全基因组、5´-UTR、Npro及E2基因序列分别进行系统发育分析和相似性比对,对分离毒株关键蛋白Npro和E2进行生物信息学分析,并进一步了解分离毒株基因组同义密码子使用模式.[结果]BVDV阳性血清样本接种MDBK细胞盲传 5代后未观察到典型的细胞病变情况,分离获得 1株非细胞病变型(NCP)BVDV毒株,命名为BVDV 24GS-19,提交NCBI(登录号:PV806860.1).IFA结果显示,分离株接种细胞后可观察到特异性红色荧光.相似性比对显示,分离毒株与BVDV CH0513株(登录号:MH901135.1)相似性达97.02%;遗传进化分析表明该毒株为BVDV-1b型.生物信息学分析显示,分离株Npro和E2蛋白不稳定指数分别为49.11和57.61,且存在氨基酸突变位点和磷酸化位点;E2蛋白存在N-糖基化和C-甘露糖基化位点;E2蛋白中预测到 2个抗原表位(第 250-269、687-706位氨基酸处).同义密码子使用模式分析显示,该毒株对脯氨酸、精氨酸、丙氨酸和苏氨酸的同义密码子使用存在偏好性.[结论]本研究分离获得 1株NCP型BVDV流行毒株,经基因组和遗传演化分析鉴定为BVDV-1b亚型,其与分离株BVDV CH0513相似性较高.本试验结果为甘肃地区牛病毒性腹泻的防控和疫苗研发提供了数据支撑.

[Objective]The purpose of this experiment was to understand the prevalence and molecular characteristics of Bovine viral diarrhea virus(BVDV)in cattle herds in Gansu,and provide a reference for BVDV prevention and control in this area.[Method]Virus isolation was performed on previously identified BVDV-positive serum samples.The isolated viral strains were identified using an indirect immunofluorescence assay(IFA)with a monoclonal antibody targeting the BVDV NS3 protein.Based on the whole genome sequencing data of the isolates,phylogenetic and similarity comparison were conducted on the whole genome,5´-UTR,Npro,and E2 gene sequences.Bioinformatic analysis was performed on the key proteins Npro and E2 of the isolated strain.Additionally,the synonymous codon usage pattern of the isolate genome was analyzed.[Result]After inoculating BVDV-positive serum samples into MDBK cells and conducting blind passage for 5 generations,no typical cytopathic effects were observed.One non-cytopathic(NCP)BVDV strain was isolated and named BVDV 24GS-19,then submited to NCBI,and the accession No.:PV806860.1.IFA results showed that the isolated strain could be observed to produce specific red fluorescence when inoculated into cells.The similarity comparison showed that the isolated strain was 97.02%similar to the BVDV CH0513 strain(accession No.:MH901135.1),and genetic evolution analysis indicated that the strain belonge to the BVDV-1b type.Bioinformatics analysis revealed that the instability indices of Npro and E2 proteins of the isolated stranin were 49.11 and 57.61 respectively,and there were amino acid mutation sites and phosphorylation sites.E2 protein had N-glycosylation and C-mannosylation sites,and had two antigenic epitopes(250-269 and 687-706 amino acids).Analysis of the synonymous codon usage pattern revealed a preference for codons encoding proline,arginine,alanine,and threonine.[Conclusion]In this study,one NCP-type BVDV epidemic strain was isolated.Through genomic and genetic evolution analysis,it was identified as subtype BVDV-1b.It showed a high similarity to the isolated strain BVDV CH0513.This results provided data support for the prevention and control of bovine viral diarrhea in Gansu as well as the development of vaccines.
