[目的]探究鸡源B亚型禽偏肺病毒(Avian metapneuovirus subtype B,aMPV/B)对火鸡的致病性.[方法]利用前期分离获得的鸡源aMPV/B LN16株,采用滴鼻方式单次感染7周龄火鸡(200 μL,感染剂量为5×104 TCID50/只),对照组以同等方式接种等体积PBS,每组13只,试验周期为21 d.攻毒后1~7 d观察火鸡的临床症状并采集火鸡腭裂拭子,利用实时荧光定量PCR检测腭裂拭子的病毒拷贝数;攻毒后第7天每组选取3只火鸡进行剖杀,采集肺脏、气管、喉头及鼻甲骨,检测病毒的组织分布情况,利用苏木精-伊红(HE)染色观察组织器官病理学变化;攻毒后7、14和21 d分别通过翅下静脉采血,利用ELISA方法测定火鸡血清抗体水平.[结果]临床症状观察显示,攻毒后第3天,感染组火鸡开始出现流鼻涕症状,连续观察7 d后发病率为84.6%(11/13);对照组火鸡未见临床症状.实时荧光定量PCR结果显示,感染组火鸡在感染后1~7 d排毒,第3天达到高峰,平均病毒拷贝数达3.7×105 拷贝/200 μL;对照组火鸡均为阴性.病毒组织分布检测结果显示,攻毒后第7天,感染组火鸡鼻甲骨和气管病毒检出率为33.3%(1/3),肺脏和喉头组织未检测到病毒.组织病理学结果显示,感染组火鸡的鼻甲骨、肺脏、气管均表现出不同程度的病理损伤,主要表现为炎性细胞浸润;对照组未见明显病理变化.ELISA结果显示,感染后第7天血清样品全部呈阳性,第14天aMPV平均抗体水平为8 934,达到峰值.[结论]本研究表明鸡源aMPV/B对火鸡具有致病性,试验结果为后续火鸡禽偏肺病毒病的临床诊断和防控提供了重要参考.

[Objective]This study aimed to investigate the pathogenicity of Avian metapneuovirus subtype B(aMPV/B)from chickens to turkeys.[Method]The previously isolated aMPV/B LN16 strain of chicken origin was used for a single nasal drip infection of 7-week-old turkeys(with 200 μL containing a dose of 5×104 TCID50 per bird.Turkeys in control group were inoculated with the same volume of PBS in the same way.13 in each group,and the trial period was 21 days.From 1 to 7 days after challenge,the clinical symptoms of turkeys were observed,and the palatal cleft swabs were collected.Real-time quantitative PCR was used to detect the viral copy numbers in the palatal cleft swabs.On the 7th day after challenge,three turkeys from each group were euthanized and necropsied.The lungs,trachea,larynx,and nasal turbinates were collected to examine the viral tissue distribution.The pathological changes of tissues and organs were observed by hematoxylin-eosin(HE)staining.Additionally,blood samples were collected from the brachial vein at 7,14,and 21 days after challenge.The antibody levels in turkey serum were determined by ELISA method.[Result]Clinical observation revealed that turkeys in infection group developed nasal discharge starting at 3 days after challenge.After continuous observation for 7 days,the incidence rate was 84.6%(11/13).No clinical symptoms were observed of turkeys in control group.Real-time quantitative PCR demonstrated turkeys in infection group excreted the virus 1 to 7 days after challenge,reaching the peak on the 3rd day,with an average virus copy number of 3.7×105 copies/200 μL.All the turkeys in control group were negative.Viral tissue distribution analysis showed that on the 7th day after challenge,the detection rate of virus in nasal conchae bone and trachea of the infected turkeys was 33.3%(1/3),while no virus was detected in lung and laryngeal tissues.Histopathological results showed that the nasal conchae,lungs,and trachea of turkeys in infection group all exhibited pathological damage of varying degrees,mainly manifested as inflammatory cell infiltration.No obvious pathological changes were observed in control group.ELISA results indicated that all serum samples were positive on the 7th day after challenge,and the average antibody reached peak levels at 8 934 at 14 days after challenge.[Conclusion]This study indicated that chicken aMPV/B was pathogenic to turkeys.The test results provided an important reference for the subsequent clinical diagnosis and prevention and control of avian metapneumovirus disease in turkeys.
