本试验利用前期从小麦田土壤样品中筛选到的一株对DON有高效降解能力的菌株JM-3,采用PacBio第三代测序技术对该菌株进行了全基因组测序,并利用HGAP、Canu、CGView、Glimmer、KOBAS、MEGA、Mauve等软件对该基因组进行了组装与分析、圈图的绘制、编码基因的预测与功能注释、系统进化分析、比较基因组学分析等.全基因组分析和基因预测结果表明,JM-3 基因组全长约 4.7 Mb,GC含量为 63.9%,共预测出了 4534 个编码基因.通过GO、COG、KEGG、CAZy等数据库比对,分别注释了 2217、3304、2256、478 个基因.系统进化分析表明,JM-3 菌株与德沃斯氏菌的进化关系最近,确定其为德沃斯氏菌(Devosia sp.).比较基因组学分析结果表明,尽管JM-3菌株与Devosia sp.A16 和Devosia sp.D6-9 均具有良好的DON降解能力,但其基因组存在多个基因片段插入/缺失、重排和易位的现象,表明JM-3 是一种新的DON降解菌株.本研究对进一步挖掘JM-3 基因组中的DON降解关键酶提供了数据支撑和科学依据.

In this experiment a strain of bacteria JM-3 from soil samples in wheat fields and with high efficiency in degrading deoxynivalenol(DON)were used.The third-generation sequencing technology was used to measure the whole genome sequence of the JM-3 strain.The genome was assembled,analyzed,and circle mapped using software such as HGAP,Canu,CGView.In addition,coding gene prediction,functional annotation,systematic evolution analysis,and collinearity analysis were performed using software of Glimmer,KOBAS,MEGA,and Mauve.The whole genome analysis results showed that the JM-3 genome was approximately 4.7 Mb in length,with a GC content of 63.9%.4534 coding genes were predicted.By comparing GO,COG,KEGG,and CAZy databases,2217,3304,2256,and 478 genes were annotated,respectively.Systematic evolutionary analysis showed that the JM-3 strain had the closest evolutionary relationship with Devosia bacteria.Based on our previous morphological,physiological,and biochemical analysis results,it was identified as Devosia sp.The results of comparative genome analysis indicated that,although JM-3 strain had significant DON degradation ability as well as previously reported Devosia sp.A16 and Devosia sp.D6-9,its genome existed multiple instances of gene segment insertion/deletion,rearrangement,and translocation,indicating that JM-3 was a new DON degrading strain.These results provide important data for further exploration of DON degrading enzymes from the JM-3 genome.
