鸡舍养殖模式与优质肉鸡肠道形态及微生物群落的关联OA
Association between housing modes of chicken coops and intestinal morphology as well as microbial community of high quality broilers
为明确不同类型鸡舍中优质肉鸡的肠道形态与微生物群落变化,选取健康的1日龄L系优质肉鸡公鸡240只,随机分成2组,饲养于密闭式环控舱(Ⅰ组)和普通半封闭式鸡舍(Ⅱ组).第15周龄时选取2组公鸡各24只屠宰进行肠道形态的测定,利用16S rDNA测序分析盲肠内容物中微生物多样性.结果表明:1)15周龄时,Ⅰ组相比于Ⅱ组显著提升了饲料转化率(P<0.05),同时十二指肠、空肠和回肠的绒毛高度也显著提升、隐窝深度显著降低(P<0.05).2)2组盲肠肠道微生物Beta多样性存在显著差异表明Ⅰ组中富集了激活脂内代谢与增强肠道屏障的菌群,如双歧杆菌属(Bifidobacterium)、放线菌门(Actinobacteriota)和疣微菌门(Verrucomicrobiota),Ⅱ组富集了抑制相关酶活性与阻碍肠上皮能量代谢的拟杆菌门(Bacteroidota)、巴氏菌科(Barnesiellaceae)和脱硫杆菌门(Desulfobacterota)(P<0.05).综上,在不同鸡舍环境参数调控模式下,优质肉鸡的肠道组织形态与微生物群落结构存在明显差异,其中密闭式环控舱模式更有利于促进肠道健康发育.保持肠道内环境稳态不仅对集约化养殖中的生产挑战具有重要实践价值,也为开展优质肉鸡在精细化调控条件下的微生物群落组学研究提供重要参考.
To clarify the intestinal morphology and microbiome changes of high-quality broilers in different types of chicken houses,240 healthy 1-day-old high-quality broilers of the L line were selected and randomly divided into two groups,which were raised in a closed environmental control cabin(Group Ⅰ)and a common semi-closed chicken coop(Group Ⅱ).At the 15th week of age,24 roosters in each group were selected and slaughtered for intestinal morphology determination,and the microbial diversity in cecum contents was analyzed by 16S rDNA sequencing.The results showed as follows:1)At 15 weeks of age,Group Ⅰ significantly improved the feed conversion rate compared with group Ⅱ(P<0.05),the villus height of duodenum,jejunum and ileum was also significantly increased,and the crypt depth was significantly decreased(P<0.05).2)The significant differences in beta diversity of the cecal microbiota between the two groups indicated that Group Ⅰ was enriched with Bifidobacterium,Actinobacteriota and Verruco microbiota,which was capable of activating lipid metabolism and enhancing the intestinal barrier,Group Ⅱ was enriched with Bacteroidota,Barnesiellaceae and Desulfobacterota,which inhibited related enzyme activity and hinder intestinal epithelial energy metabolism(P<0.05).In summary,there are significant differences in the intestinal tissue morphology and microbial community structure of high-quality broiler chickens by environmental parameters of different types of chicken houses.Among them,the closed-loop control model is more suitable for healthy intestinal development of high-quality broilers.Maintaining a stable intestinal environment is not only of great practical value for the production challenges in intensive farming,but also provides important reference for conducting microbiome research on high-quality broiler chickens under refined regulation conditions.
