T细胞受体(TCR)与抗原肽-主要组织相容性复合物(pMHC)的相互作用是适应性细胞免疫应答的核心环节.TCR-pMHC结合不仅涉及化学作用力,如亲和力、亲合力及功能亲合力(FA),还受到机械力的动态调节,从而影响其结合构象和信号转导.因此,T细胞抗原识别和激活过程与化学作用力及机械力均密切相关.在病毒感染中,TCR-pMHC间的作用力差异对不同病原体或感染状态下的T细胞应答、病毒清除及免疫逃逸具有显著影响.本文重点综述TCR-pMHC结合过程中化学作用力与机械力的作用机制及其对病毒感染诱导T细胞应答的调控,如何作为传统化学作用力的补充,从化学与力学相结合的视角,为病毒感染性疾病的免疫治疗提供新的思路和策略.

Interaction between T cell receptor(TCR)and antigen peptide-major histocompatibility complex(pMHC)is a central component of adaptive cellular immune response.TCR-pMHC binding is governed not only by chemical forces,such as affinity,avidity and functional avidity(FA),but is also dynamically regulated by mechanical forces,which in turn influence binding conforma-tion and subsequent signal transduction.Therefore,process of T cell antigen recognition and activation are intricately linked to both chemical and mechanical forces.In context of viral infections,interplay of forces between TCR and pMHC exhibits distinct characteris-tics and plays differential roles in T cell responses,viral clearance and immune escape,depending on specific pathogen and infection state.This review focuses on elucidating mechanisms by which chemical and mechanical forces act in TCR-pMHC binding process and regulate T cell responses during viral infections.It further explores how mechanical forces can complement traditional chemical forces.By integrating insights from both chemical and mechanical perspectives,this review aims to provide novel ideas and strategies for immunotherapy of viral infectious diseases.
