垂穗披碱草近壳多胞叶斑病快速检测技术的建立与应用OA
Establishment and application of molecular detection for Parastagonospora nodorum-induced leaf spot disease in Elymus nutans
为建立一种由颖枯近壳多胞病菌(Parastagonospora nodorum)引发垂穗披碱草(Elymus nutans)叶斑病的快速检测体系,利用病原菌的RPB2(RNA polymerase Ⅱ subunit 2)保守序列设计了具有高度特异性的引物Pr-F/Pr-R,建立了一种高效、快速、稳定的PCR检测垂穗披碱草近壳多胞叶斑病的分子检测技术.结果表明:1)检测体系可以特异地从颖枯近壳多胞菌中扩增出一条127 bp的条带,Pr-F/Pr-R对颖枯近壳多胞菌基因组DNA的检测灵敏度为100pg/μL.2)引物Pr-F/Pr-R可对发病垂穗披碱草叶片组织中颖枯近壳多胞病菌进行特异性检测,还可用于对田间疑似带病垂穗披碱草植株的快速检测.本研究结果为垂穗披碱草叶斑病的早期发现和针对性防治提供了技术支持.
To establish a rapid detection system for leaf spot disease of Elymus nutans caused by Parastagonospora nodorum,highly specific primers Pr-F/Pr-R targeting the conserved RPB2(RNA polymerase Ⅱ subunit 2)gene sequence of the pathogen was designed.An efficient,rapid,and stable PCR molecular detection technique was developed to identify Parastagonospora leaf spot in E.nutans.The results indicated:1)The detection system specifically amplified a 127 bp band from P.nodorum,and the sensitivity of Pr-F/Pr-R for detecting genomic DNA of P.nodorum was 100 pg/μL.2)The Pr-F/Pr-R primers can specifically detect P.nodorum in infected E.nutans leaf tissues,as well as in suspected diseased field-collected E.nutans plants.These results provide a technical basis for the early detection and targeted control of leaf spot disease in E.nutans.
