基于TNF-α/NF-κB信号通路探究三橘荔核汤治疗Ⅲ型前列腺炎的机制OA
Mechanism of Sanjulihe Decoction in the treatment of type Ⅲ prostatitis based on TNF-α/NF-κB signaling pathway
目的 基于肿瘤坏死因子-α(TNF-α)/核因子-κB(NF-κB)信号通路探讨三橘荔核汤治疗Ⅲ型前列腺炎的机制.方法 将70 只SD大鼠随机分为7 组,每组10 只.正常组大鼠正常饲养,假手术组大鼠前列腺腹侧叶注射生理盐水,模型组、前列通瘀胶囊组、三橘荔核汤低剂量组、三橘荔核汤中剂量组、三橘荔核汤高剂量组大鼠前列腺腹侧叶注射大鼠前列腺蛋白提纯液与弗氏完全佐剂混合物建立Ⅲ型前列腺炎模型.造模后第3 天开始,三橘荔核汤低、中、高剂量组分别给予5.4 g/(kg·d)、10.8 g/(kg·d)、21.6 g/(kg·d)三橘荔核汤灌胃,前列通瘀胶囊组给予0.54 g/(kg·d)前列通瘀胶囊水溶剂灌胃,正常组、假手术组和模型组给予等量生理盐水灌胃,均1 次/d.连续灌胃4 周后,HE染色观察前列腺组织病理形态,ELISA法检测前列腺组织中白细胞介素-2(IL-2)、白细胞介素-18(IL-18)、转化生长因子-β1(TGF-β1)、环氧化酶-2(COX-2)含量,Western blot法和RT-PCR法检测前列腺组织中TNF-α/NF-κB信号通路相关蛋白及mRNA表达情况.结果 与假手术组和正常组比较,模型组大鼠前列腺组织中有大量炎性细胞浸润;前列腺组织中IL-2 含量均明显降低(P均<0.05),IL-18、TGF-β1、COX-2 含量和NF-κB、IKKα、TNF-α、IκBα蛋白及mRNA相对表达量均明显升高(P均<0.05).与模型组比较,各用药组大鼠前列腺组织炎性细胞浸润减少;前列腺组织中IL-2 含量均明显升高(P均<0.05),IL-18、TGF-β1、COX-2 含量和NF-κB、TNF-α、IκBα蛋白及mRNA相对表达量均明显降低(P均<0.05);与前列通瘀胶囊组比较,三橘荔核汤中、高剂量组大鼠前列腺组织中IL-2 含量均更高(P均<0.05),IL-18、TGF-β1、COX-2 含量和NF-κB、IKKα、IκBα蛋白及mRNA相对表达量均更低(P均<0.05).结论 三橘荔核汤可能通过调控TNF-α/NF-κB信号通路发挥治疗Ⅲ型前列腺炎的作用.
Objective It is to explore the mechanism of Sanjulihe Decoction in the treatment of typeⅢprostatitis based on tumor necrosis factor-α(TNF-α)/nuclear factor-κB(NF-κB)signaling pathway.Methods Seventy SD rats were ran-domly divided into 7 groups,with 10 rats in each group.The rats of the normal group were normally fed,while the rats of the sham operation group were injected with normal saline into ventral prostate,the rats of the model group,Preprostatitis Capsule group,low-dose,medium-dose,and high-dose groups of Sanjulihe Decoction were injected with a mixture of puri-fied rat prostate protein and Freund's complete adjuvant into ventral prostate to establish models of type Ⅲ prostatitis.Starting from the 3rd day after modeling,the low-dose,medium-dose,and high-dose groups of Sanjulihe Decoction were administered Sanjulihe Decoction 5.4 g/(kg·d),10.8 g/(kg·d),and 21.6 g/(kg·d)via gavage,respectively,the Preprostatitis Capsule group was givn aqueous solution of Preprostatitis Capsule 0.54 g/(kg·d)via gavage,while the nor-mal group,sham operation group and model group were given an equivalent volume of saline via gavage,all once daily.Af-ter 4 weeks of continuous gavage,the pathological morphology of prostate tissue was observed by HE staining,the contents of interleukin-2(IL-2),interleukin-18(IL-18),transforming growth factor-β1(TGF-β1),and cyclooxygenase-2(COX-2)in prostate tissue were determined by ELISA,the expression of proteins and mRNA related to the TNF-α/NF-κB signa-ling pathway in prostate tissue were detected by Western blot and RT-PCR methods.Results Compared with the sham opera-tion group and normal group,extensive inflammatory cell infiltration in prostate tissue was found in rats of the model group;the content of IL-2 in prostate tissue of the rats was significantly reduced(P<0.05),while the contents ofIL-18,TGF-β1 and COX-2,as well as the relative proteins and mRNA expressions of NF-κB,IKKα,TNF-α,IκBα were significantly in-creased(all P<0.05).Compared with the model group,inflammatory cell infiltration in prostate tissue was reduced in rats of all medicated groups;the contents of IL-2 in prostate tissue of the rats were significantly increased(all P<0.05),while the contents of IL-18,TGF-β1 and COX-2,as well as the relative proteins and mRNA expressions of NF-κB,TNF-α,IκBα were significantly decreased(all P<0.05).Compared with Preprostatitis Capsule group,the contents of IL-2 in prostate tissue were more higher(both P<0.05),while the contents ofIL-18,TGF-β1 and COX-2,as well as relative protein and mRNA expressions of NF-κB,IKKα and IκBα in prostate tissue were more lower in rats of the medium-dose,and high-dose groups of Sanjulihe Decoction(all P<0.05).Conclusion Sanjulihe Decoction can treat type Ⅲ prostatitis possibly via regulating TNF-α/NF-κB signaling pathway.
