妇康栓对产后大鼠子宫复旧的影响研究OA

目的 探讨妇康栓对产后大鼠子宫复旧的调节作用.方法 将 50 只妊娠后SD大鼠随机分为模型组、卡孕栓组、妇康栓低剂量组、妇康栓中剂量组、妇康栓高剂量组各 10 只,另取10 只未孕雌鼠作为空白组.分娩后第7 天起,卡孕栓组阴道给予0.105 mg/kg卡孕栓,妇康栓低、中、高剂量组阴道给予0.262 g/kg、0.525 g/kg、1.05 g/kg妇康栓,均1 次/d.连续干预14d后,HE染色观察子宫组织病理学形态;ELISA法检测血清性激素[雌二醇(E2)、孕酮(P)],炎症因子[肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)],免疫球蛋白[免疫球蛋白G(IgG)、免疫球蛋白A(IgA)],生长因子[转化生长因子-β1(TGF-β1)、转化生长因子-β3(TGF-β3)、血管内皮生长因子(VEGF)]水平;Western blot法检测子宫组织中基质金属蛋白酶-9(MMP-9)、基质金属蛋白酶抑制因子-1(TIMP-1)蛋白表达情况.结果 与空白组比较,模型组大鼠子宫腔形态异常,内膜较薄且连续性部分中断;血清E2、IL-10、TGF-β3、VEGF水平和TGF-β3/TGF-β1 比值均明显降低(P均<0.05),血清P、TNF-α、IL-6、IgA、IgG、TGF-β1 水平均明显升高(P均<0.05);子宫组织中MMP-9 蛋白相对表达量明显升高(P<0.05),TIMP-1 蛋白相对表达量明显降低(P<0.05).与模型组比较,各给药组大鼠子宫腔形态趋于正常,内膜连续性较好;卡孕栓组和妇康栓高剂量组大鼠血清E2、P(卡孕栓组除外)、IL-10、TGF-β3、VEGF(卡孕栓组除外)水平和TGF-β3/TGF-β1 比值均明显升高(P均<0.05),血清TNF-α、IL-6、IgG、TGF-β1水平均明显降低(P均<0.05);子宫组织中MMP-9 蛋白相对表达量明显降低(P<0.05),TIMP-1蛋白相对表达量明显升高(P<0.05).结论 妇康栓可以减轻炎症反应,调节性激素、生长因子水平和基质金属蛋白酶家族及其抑制物平衡,促进子宫基质重塑,有助于产后子宫复旧.

Objective:It is to investigate the regulatory effect of Fukang Suppository on uterine involution in postpartum rats.Methods Fifty pregnant SD rats were randomly divided into model group,Carboprost Suppository group,and low-dose,medium-dose,and high-dose groups of Fukang Suppository,with 10 rats in each group,another 10 non-pregnant fe-male rats were taken as the blank group.Starting from the 7th day after delivery,the Carboprost Suppository group was giv-en Carboprost Suppository 0.105 mg/kg by vaginal administration,the low-dose,medium-dose and high-dose groups of Fu-kang Suppository were given Fukang Suppository 0.262 g/kg,0.525 g/kg,and 1.05 g/kg by vaginal administration,re-spectively,all administrated once daily.After 14 days of continuous treatment,the histopathology of uterine tissue was ob-served by HE staining;the serum levels of sex hormones[estradiol(E2),progesterone(P)],inflammatory cytokines[tumor necrosis factor-α(TNF-α),interleukin-6(IL-6),interleukin-10(IL-10)],immunoglobulins[immunoglobulin G(IgG),immunoglobulin A(IgA)],and growth factors[transforming growth factor-β1(TGF-β1),transforming growth fac-tor-β3(TGF-β3),vascular endothelial growth factor(VEGF)]were measured by ELISA;the protein expressions of matrix metalloproteinase-9(MMP-9)and tissue inhibitor of metalloproteinase-1(TIMP-1)in uterine tissue were detected by Western blot analysis.Results Compared with the blank group,the uterine cavity morphology of rats in the model group was abnormal,with thinner endometrium and partial discontinuity of endometrial continuity;the serum levels of E2,IL-10,TGF-β3,and VEGF,as well as the TGF-β3/TGF-β1 ratio in the rats were significantly reduced(all P<0.05),while the serum levels of P,TNF-α,IL-6,IgA,IgG and TGF-β1 were significantly increased(all P<0.05);the relative protein expression of MMP-9 in uterine tissue was significantly increased(P<0.05),while the relative protein expression of TIMP-1 was significantly reduced(P<0.05).Compared with the model group,the uterine cavity morphology of rats in each medicated group was approaching normal,with better endometrial continuity;the serum levels of E2,P(except for the Carboprost Suppository group),IL-10,TGF-β3,and VEGF(except for the Carboprost Suppository group),as well as the TGF-β3/TGF-β1 ratio in rats of the Carboprost Suppository group and high-dose group of Fukang Suppository were sig-nificantly increased(all P<0.05),while the serum levels ofTNF-α,IL-6,IgG and TGF-β1 were significantly decreased(all P<0.05);the relative protein expression of MMP-9 in uterine tissue was significantly decreased(P<0.05),while the relative protein expression of TIMP-1 was significantly increased(P<0.05).Conclusion Fukang Suppository can alle-viate inflammatory response,regulate the levels of sex hormones and growth factors and balance of matrix metalloproteinase family and their inhibitors,promote the remodeling of uterine matrix,and contribute to uterine involution in postpartum rats.
