新葛根芩连汤调控BDNF/TrkB信号通路和抑制铁死亡防治阿尔茨海默病的研究OA

目的 探讨新葛根芩连汤通过调控脑源性神经营养因子(BDNF)/酪氨酸激酶B(TrkB)信号通路和抑制铁死亡对阿尔茨海默病(AD)的干预效应.方法 取90 只SPF级雄性SD大鼠,随机选择10只作为空白组,其余80只大鼠采用慢性铝过负荷方式建立AD模型.将旷场实验筛选出的60 只AD大鼠随机分为5 组.新葛根芩连汤高、中、低剂量组分别给予725 mg/mL、363 mg/mL、183 mg/mL新葛根芩连汤灌胃,多奈哌齐组给予0.5 mg/kg多奈哌齐灌胃,空白组和模型组给予等体积纯水灌胃,均 1 次/d.连续灌胃 5 周后,Morris水迷宫实验检测大鼠学习记忆能力,尼氏染色观察海马CA1、CA3、DG区神经元病理形态,酶联免疫吸附法检测血清β淀粉样蛋白1-42(Aβ1-42)、Fe2+含量及海马组织中Aβ1-42 含量,组织铁含量试剂盒检测海马组织中Fe2+含量,免疫组化法检测海马CA1、CA3、DG区花生四烯酸5-脂氧合酶(5-LOX)蛋白表达情况,West-ern blot法检测海马组织中BDNF、p-TrkB蛋白表达情况.结果 与空白组比较,模型组大鼠逃避潜伏期明显延长(P<0.05),平台穿越次数明显减少(P<0.05);海马CA1、CA3、DG区神经元计数明显减少(P均<0.05);血清和海马组织中Aβ1-42、Fe2+含量均明显升高(P均<0.05);海马CA1、CA3、DG区5-LOX蛋白表达平均光密度值均明显升高(P均<0.05);海马组织中BDNF、p-TrkB蛋白相对表达量均明显降低(P均<0.05).与模型组比较,各药物组大鼠逃避潜伏期均明显缩短(P均<0.05),海马CA1、CA3、DG区5-LOX蛋白表达平均光密度值均明显降低(P均<0.05),海马组织中BDNF蛋白相对表达量均明显升高(P均<0.05);新葛根芩连汤高剂量组和多奈哌齐组大鼠平台穿越次数均明显增多(P均<0.05);新葛根芩连汤中、高剂量组和多奈哌齐组大鼠海马CA1、CA3、DG区神经元计数明显增多(P均<0.05),血清和海马组织中Aβ1-42、Fe2+含量均明显降低(P均<0.05),海马组织中p-TrkB蛋白相对表达量均明显升高(P均<0.05).结论 新葛根芩连汤可通过调控BDNF/TrkB和抑制铁死亡,进而改善AD大鼠学习记忆能力.

Objective It is to explore the intervention effect of new Gegen Qinlian Decoction(NGQD)on Alzheimer's disease(AD)via regulating brain-derived neurotrophic factor(BDNF)and tyrosine kinase B(TrkB)and inhibiting fer-roptosis.Methods Ninty male SD rats of SPF grade were taken,10 of which were randomly selected as blank group,and the other 80 rats were prepared by by chronic aluminum overload mode to establish AD models.60 AD rats screened by open field test were randomly divided into 5 groups.The high-dose,medium-dose,and low-dose groups of NGQD were giv-en NGQD 725 mg/mL,363 mg/mL,and 183 mg/mL by gavage,respectively,the donepezil group was given donepezil 0.5 mg/kg by gavage,the blank and model groups were given an equal volume of pure water by gavage,all administered once daily.After 5 weeks of continuous administration,the learning and memory abilities of the rats were assessed by Mor-ris water maze test,the pathological morphology of neurons in hippocampal CA1,CA3,and DG regions were observed by Nissl staining,the contents of β-amyloid 1-42(Aβ1-42)and Fe2+in serum,and the content of Aβ1-42 in hippocampal tissue were measured by ELISA,the content of Fe2+in hippocampal tissue was measured by a tissue iron assay kit,the protein expressions of 5-lipoxygenase(5-LOX)in hippocampal CA1,CA3,and DG regions were detected by Immunohistochemis-try,and the protein expressions of BDNF and p-TrkB in hippocampal tissue were detected by Western blot analysis.Re-sults Compared with the blank group,the escape latency of rats in the model group was significantly prolonged(P<0.05),the number of times of platform crossings was significantly decreased(P<0.05);the neuron counts in hippocam-pal CA1,CA3,and DG regions were significantly reduced(all P<0.05);the contents of Aβ1-42 and Fe2+in serum and hippocampal tissue were significantly increased(all P<0.05);the mean optical density values of protein expressions of 5-LOX in hippocampal CA1,CA3,and DG regions were significantly increased(all P<0.05);the relative protein expres-sions of BDNF and p-TrkB in hippocampal tissue were significantly reduced(all P<0.05).Compared with the model group,the escape latencies of rats in the medicated groups were significantly shortened(all P<0.05),the mean optical density values of protein expressions of 5-LOX in hippocampal CA1,CA3,and DG regions were significantly reduced(all P<0.05),while the relative protein expressions of BDNF in hippocampal tissue were significantly increased(all P<0.05);the platform crossing times of rats in the high-dose group of NGQD and donepezil group were significantly increased(all P<0.05);the neuronal counts in hippocampal CA1,CA3,and DG regions of rats in the high-dose,medium dose group of NGQD and donepezil group were significantly increased(all P<0.05),the contents of Aβ1-42 and Fe2+in serum and hippocampal tissue were significantly decreased(all P<0.05),and the relative protein expression of p-TrkB in hipp-ocampal tissue was significantly increased(P<0.05).Conclusion NGQD can improve the learning and memory abilities of AD rats via regulating BDNF/TrkB signal pathway and inhibiting ferroptosis.
