电针对帕金森病小鼠中脑黑质SIRT3/NF-κB/MAPK信号通路的影响OA
Effects of electroacupuncture on the SIRT3/NF-κB/MAPK signaling pathway in the substantia nigra of mice with Parkinson's disease
目的:观察电针"风府""太冲"对帕金森病(PD)小鼠中脑黑质酪氨酸羟化酶(TH)、小胶质细胞(MG)及沉默信息调节因子3(SIRT3)/核因子κB(NF-κB)/丝裂原活化蛋白激酶(MAPK)信号通路相关蛋白的影响,探讨电针治疗PD的作用机制.方法:C57BL/6J小鼠随机分为对照组、模型组、非经穴组、电针组,每组12只.采用1-甲基-4-苯基-1,2,3,6-四氢吡啶连续腹腔注射7 d构建PD模型,电针组电针"风府""太冲";非经穴组针刺"风府""太冲"旁开3 mm并夹持电极,不予通电,两组均15 min/次,每日1次,连续14 d.采用爬杆实验和悬挂实验检测小鼠运动能力;ELISA法检测中脑黑质肿瘤坏死因子α(TNF-α)、白细胞介素(IL)-1β含量;免疫组织化学法检测小鼠中脑黑质TH阳性表达;免疫荧光标记法检测中脑黑质激活的MG阳性细胞数量;Western blot法检测小鼠中脑黑质SIRT3、NF-κB p65、p38 MAPK蛋白表达水平.结果:与对照组比较,模型组小鼠爬杆实验时间明显延长(P<0.01),悬挂实验评分明显降低(P<0.01),中脑黑质TH平均吸光度值明显降低(P<0.01),MG阳性细胞数增加(P<0.01),SIRT3 蛋白表达水平降低(P<0.01),NF-κB p65、p38 MAPK蛋白表达水平及血清TNF-α、IL-1β含量升高(P<0.01).与模型组和非经穴组比较,电针组小鼠爬杆实验时间缩短(P<0.01),悬挂实验评分升高(P<0.01),中脑黑质中TH平均吸光度值增加(P<0.01),MG阳性细胞数减少(P<0.01),SIRT3蛋白表达水平升高(P<0.05),NF-κB p65、p38 MAPK蛋白表达水平及血清TNF-α、IL-1β含量均降低(P<0.01).结论:电针可改善PD小鼠运动功能障碍,减少中脑黑质多巴胺能神经元的丢失,提高TH表达水平,其作用机制可能与调控SIRT3/NF-κB/MAPK信号通路抑制神经炎性反应有关.
Objective To investigate the effects of electroacupuncture(EA)at"Fengfu"(GV16)and"Taichong"(LR3)on tyrosine hydroxylase(TH),microglia(MG),and the silent information regulator 3(SIRT3)/nuclear factor-kappa B(NF-κB)/mitogen-activated protein kinase(MAPK)signaling pathway-related proteins in the substantia nigra of Parkinson's disease(PD)mice,and to explore the potential mechanisms underlying the therapeutic effects of EA on PD.Methods C57BL/6J mice were randomly divided into 4 groups:control group,model group,non-acupoint group,and EA group,with 12 mice in each group.A PD model was established by intraperitoneal injections of MPTP for 7 consecutive days.The EA group received EA at GV16 and LR3.The non-acupoint group underwent needle insertion 3 mm lateral to GV16 and LR3 without electrical stimulation.Treatments were administered for 15 min per session,once daily for 14 days.Motor function was assessed using the pole test and suspension test.Immunohistochemistry was used to detect TH-positive expression in the substantia nigra.Immunofluorescence was employed to quantify activated MG.Western blot was used to measure the protein expression levels of SIRT3,NF-κB p65,and p38 MAPK.ELISA was used to determine the serum levels of tumor necrosis factor-α(TNF-α)and interleukin-1β(IL-1β).Results Compared with the control group,the model group exhibited a longer total time in the pole test(P<0.01),a lower score in the suspension test(P<0.01),a reduce in mean TH optical density in the substantia nigra(P<0.01),an increase in the number of MG-positive cells(P<0.01),a decrease in SIRT3 protein expression(P<0.01).The protein expression levels of NF-κB p65 and p38 MAPK proteins in the substantia nigra,and the serum levels of TNF-α,and IL-1β were all elevated(P<0.01).Compared with model group and the non-acupoint group,the EA group showed a shortened total time in the pole test(P<0.01),a higher score in the suspension test(P<0.01),an increase in mean TH optical density in the substantia nigra(P<0.01),a decrease in the number of MG-positive cells(P<0.01),an increase in SIRT3 protein expression(P<0.05).The expression levels of NF-κB p65 and p38 MAPK proteins in the substantia nigra,and the serum contents of TNF-α and IL-1β were all decreased(P<0.01).Conclusion EA can improve motor dysfunction in PD mice,reduce the loss of dopaminergic neurons,and increase TH expression in the substantia nigra.The underlying mechanism may involve the modulation of the SIRT3/NF-κB/MAPK signaling pathway to suppress neuroinflammation.
