[目的]探明马家柚[Citrus maxima(L.)Osbeck cv.'Majiayou']线粒体基因组结构特征及系统发育关系.[方法]利用高通量测序技术对马家柚线粒体基因组进行测序,并利用生物信息学相关软件对该线粒体基因组进行组装、注释、图谱绘制以及序列特征分析,并基于线粒体基因组构建柑橘属5个物种的系统发育树.[结果]马家柚线粒体基因组大小为516 383 bp,呈完整单环状结构,GC含量为45.15%;注释到65个基因,其中包括35个编码蛋白基因、27个tRNA基因和3个rRNA基因.马家柚线粒体基因组共鉴定出64个SSR(A/T单核苷酸重复占主导地位)和100个Longrepeat序列(51个正向重复和49个回文重复).马家柚线粒体基因组的密码子偏性较弱,偏好以A、U结尾.马家柚线粒体基因组密码子使用偏好性受内部突变和自然选择的双重影响,其中自然选择对密码子使用偏好性的影响更大.马家柚线粒体基因组的最优密码子为22个(AAU、GAU、UGU、CAA、AAA、UUU、UAU、AUU、AUG、UAA、GGA、GGU、CCA、CCU、ACU、GUA、GUU、CGA、CGU、UUA、UCA和UCU),多以A、U结尾.马家柚线粒体基因组与甜橙线粒体基因组的共线性较高,两者的亲缘关系较近.马家柚有21个叶绿体基因片段转移到线粒体基因组,转移片段的总长度为12 195 bp.[结论]探明了马家柚线粒体基因组序列特征和密码子偏好性;马家柚与甜橙的遗传关系最近.

[Objective]The study aims to investigate the structural characteristics and phylogenetic re-lationships of the mitochondrial genome in Citrus maxima(L.)Osbeck cv.'Majiayou'.[Method]The mitochondrial genome of C.maxima(L.)Osbeck cv.'Majiayou'was sequenced using high-throughput sequencing.Bioinformatics software was employed to assemble,annotate,map,and analyze sequence cha-racteristics of the mitochondrial genome.A phylogenetic tree of five Citrus species was constructed based on this mitochondrial genome sequence.[Result]The mitochondrial genome size of C.maxima(L.)Os-beck cv.'Majiayou'was 516 383 bp,with a complete monocyclic structure and a GC content of 45.15%.Annotation identified 65 genes,including 35 protein coding genes,27 tRNA genes,and 3 rRNA genes.A to-tal of 64 SSRs(A/T single nucleotide repeats dominant)and 100 Longrepeat sequences(51 direct repeats and 49 palindromic repeats)were identified in the mitochondrial genome of C.maxima(L.)Osbeck cv.'Majiayou'.This mitogenome exhibited weak codon usage bias with a preference for A and U endings.The codon usage preference of C.maxima(L.)Osbeck cv.'Majiayou'mitochondrial genome was influenced by both internal mutations and natural selection,with the latter being dominant.Twenty-two optimal co-dons were determined(AAU,GAU,UGU,CAA,AAA,UUU,UAU,AUU,AUG,UAA,GGA,GGU,CCA,CCU,ACU,GUA,GUU,CGA,CGU,UUA,UCA,and UCU),all ending in A and U.The mitochon-drial genome of C.maxima(L.)Osbeck cv.'Majiayou'showed high collinearity with the mitochondrial genome of Citrus sinensis,indicating a close genetic relationship between the two.There were 21 chloro-plast gene fragments of C.maxima(L.)Osbeck cv.'Majiayou'transferred to its mitochondrial genome,with a total length of 12 195 bp.[Conclusion]Sequence characteristics and codon usage bias of mitochon-drial genome of Citrus maxima(L.)Osbeck cv.'Majiayou'were elucidated,with the closest genetic rela-tionship to Citrus sinensis among the studied Citrus species being demonstrated.
