苹果NF-X1基因家族及盐胁迫下的组织表达模式分析OA
Analysis of the NF-X1 gene family in apple and its tissue expression pattern under salt stress
[目的]以苹果转录因子NF-X1基因家族为研究对象,对其不同组织表达特性及对盐胁迫的响应规律进行研究,为进一步探索苹果砧木抗盐功能提供参考.[方法]以苹果砧木八棱海棠(Malus×robusta)为供试材料,利用生物信息学方法,对NF-X1基因家族成员进行鉴定、基本理化性质分析、亚细胞定位预测及系统进化分析;使用TBtools,对MdNF-X1基因家族成员的基因结构、保守结构域、保守基序、启动子顺式元件等进行可视化分析;采用实时荧光定量PCR(RT-qPCR)技术,检测MdNF-X1基因家族的组织表达特异性及盐胁迫下不同组织中的响应特性.[结果]苹果MdNF-X1基因家族有MdNF-X1-1、MdNF-X1-2、MdNF-X1-3、MdNF-X1-4共4个成员,编码蛋白分别含1 162,1 138,865和830个氨基酸,亚细胞定位预测均为细胞核内蛋白.基于氨基酸序列的系统进化分析显示,4个 MdNF-X1成员可分为2个亚组,且均与葡萄NF-X1家族亲缘关系最近.蛋白功能域分析结果显示,MdNF-X1家族成员均含有NF-X1-zinc-finger保守结构域,属于锌指转录因子;顺式作用元件分析表明,MdNF-X1基因家族启动子中含有盐、干旱胁迫及脱落酸、生长素等响应元件.RT-qPCR检测显示,MdNF-X1-1和MdNF-X1-3在根中表达量较高,MdNF-X1-2在根和叶中表达较高且表达量相近,MdNF-X1-4在各个组织中表达量均较低;在盐胁迫处理下,4个MdNF-X1基因在根、茎、叶中对盐胁迫均有响应,其中以MdNF-X1-1基因在根系中对盐胁迫的响应最为显著,相对表达量高于MdNF-X1基因家族的其他成员.[结论]苹果砧木八棱海棠中的MdNF-X1-1基因在盐胁迫下显著诱导表达,表明其可能参与八棱海棠对盐胁迫的应答调控,为进一步揭示其在抗盐胁迫中的具体功能提供了依据.
[Objective]The apple transcription factor NF-X1 gene family was used as the research ob-ject to study its expression characteristics in different tissues and the response pattern to salt stress,provi-ding a reference for further research on the salt resistance function of apple rootstocks.[Method]Using the apple rootstock,Malus×robusta,as the test material,bioinformatics methods were used to identify members of the NF-X1 gene family,analyze the basic physicochemical properties,predict the subcellular localization,and perform the phylogeny analysis.The gene structures,conserved domains,conserved mo-tifs,and promoter cis-elements of the MdNF-X1 gene family members were visualized using TBtools.Re-al-time quantitative PCR(RT-qPCR)was used to detect the tissue-specific expression patterns of the MdNF-X1 gene family and its response properties in different tissues under salt stress.[Result]The apple MdNF-X1 gene family comprises four members,MdNF-X1-1,MdNF-X1-2,MdNF-X1-3,and MdNF-X1-4,enco-ding proteins with 1 162,1 138,865 and 830 amino acids,respectively,all predicted to be nuclear in subcel-lular localization.Phylogenetic analysis based on amino acid sequences revealed that the four MdNF-X1 members could be classified into two subgroups,all being most closely related to the grape NF-X1 family.Protein functional domain analysis showed that all MdNF-X1 family members contained the NF-X1-zinc-finger conserved domain,identifying them as zinc-finger transcription factors.Cis-acting element analysis demonstrated that the promoters of MdNF-X1 gene family contained response elements to salt stress,drought stress and response elements including abscisic acid and auxin.RT-qPCR analysis showed that MdNF-X1-1 and MdNF-X1-3 had higher expression in roots;NF-X1-2 had higher and similar expression in roots and lea-ves;MdNF-X1-4 had lower expression in all tissues.Under salt stress,all four MdNF-X1 genes respon-ded in roots,stems,and leaves.Among them,MdNF-X1-1 exhibited the most significant response to salt stress in the roots,with its relative expression exceeding other family members.[Conclusion]The expres-sion of MdNF-X1-1 gene in the apple rootstock Malus×robusta was significantly induced under salt stress,suggesting its involvement in the response regulation of Malus×robusta under salt stress.This pro-vides a basis for further revealing its specific functions in salt stress resistance.
