pH/温度双响应壳聚糖基可注射水凝胶的制备及性能OA
Preparation and properties of pH/temperature responsive chitosan based injectable hydrogels
响应型可注射水凝胶作为一种能感知外界环境变化并作出响应的高分子材料,以溶液状态注射,在作用部位发生相转变形成具有三维网络结构的凝胶,具有生物相容性以及与天然组织相似性等特点,广泛应用于药物递送、组织工程等医学领域.为了实现"原位成胶-精准响应-按需释药"协同功能,以聚阳离子壳聚糖(CS)为基体,温敏型阴离子聚合物(PNAM)以及碳酸氢钠(SHC)/甘油磷酸钠(GP)作为交联剂,通过双离子交联法分别构建了2种pH/温度双响应的可注射水凝胶,同时探究了单因素(浓度、配比等)对凝胶化时间的影响,通过红外光谱和扫描电镜对其结构和形貌进行了表征,重点评估了材料溶胀性和生物相容性.结果表明,当CS质量浓度为20 mg/mL、PNAM质量浓度为4 mg/mL,与SHC三者配比为10∶2∶5时,37℃下CS-PNAM-SHC水凝胶成胶时间为180 s,该水凝胶呈现出多孔表面且溶胀速率较高,可以实现缓慢、控制释放药物,可用于细胞或药物载体;当CS质量浓度为20 mg/mL、PNAM质量浓度为3 mg/mL,与GP配比为10∶13∶1时,CS-PNAM-GP水凝胶成胶时间为90 s,该水凝胶表现为层状结构,溶胀速度缓慢,可应用于制备支架材料.2种水凝胶均具有良好的生物相容性,且在人体37℃时做出智能响应,在生物医学领域具有一定的应用前景.
Stimuli-responsive injectable hydrogel,a kind of polymer material that can respond to changes in the external environment,is injected in solution form,and forms gel with three-dimen-sional network structure after phase transition at the site of action.It has the characteristics of biocompatibility and similarity with natural tissue,which is widely used in medicine fields such as drug delivery and tissue engineering.To realize the synergistic function of"in-situ gelation-precise response-on-demand drug release",two injectable hydrogels with pH/temperature dual response were constructed by using polycationic chitosan(CS)as the matrix,temperature-sensitive anionic polymer(PNAM),and sodium bicarbonate(SHC)/sodium glycerophosphate(GP)as crosslink-ing agents through the dual-ion crosslinking method.The influence of single factors(concentra-tion,ratio,etc.)on the gelation time was investigated,and their structures and morphologies were characterized by using infrared spectroscopy and scanning electron microscopy.The swelling property and biocompatibility were explored in detail.The results showed that when the concen-tration of chitosan(CS)was 20 mg/mL,temperature-sensitive polymer(PNAM)was 4.0 mg/mL,and the ratio was 10∶2∶5,the gelation time of CS-PNAM-SHC hydrogel was 180 s at 37℃.The hydrogel presents a porous surface and a high swelling rate,which can realize the con-trolled and sustain release of drugs.When the concentration of CS was 20 mg/mL,the concentra-tion of PNAM was 3.0 mg/mL,and the ratio was 10∶13∶1,the formation time of CS-PNAM-GP hydrogel was 90 s,and the hydrogel showed a stratified structure with slow swelling speed,which can be used in the preparation of scaffold materials.Both hydrogels have good biocompati-bility,and both can respond intelligently at 37℃,which are expected to have wide application prospect in the biomedical field.
