桃胶缓解重金属所致皮肤成纤维细胞损伤的作用研究OA
Protective effects of peach gum on heavy metal-induced damage in human skin fibroblasts
构建了由硫酸镍(NiSO4)诱导人皮肤成纤维细胞(HFF-1)的重金属损伤模型,通过细胞存活率、形态学观察及DNA损伤标志物(γ-H2AX)检测系统评估了桃胶对重金属诱导的人皮肤成纤维细胞损伤的保护作用.研究结果显示,NiSO4(8 mmol/L)暴露12 h使HFF-1细胞存活率降至44.65%(p<0.01),与对照组相比,γ-H2AX在细胞中的表达水平显著升高至27.16倍;经8 mg/mL 桃胶预处理的细胞存活率与模型组相比显著升高,为59.22%(p<0.01),细胞中γ-H2AX表达水平相比模型组显著降低,为对照组的3.8倍.通过组分分析显示,桃胶总糖含量为43.8%(苯酚-硫酸法测定),其多糖组分平均分子量为803.81 kDa(高效液相凝胶色谱测定),单糖组成主要为阿拉伯糖(47.96%)、半乳糖(33.16%)、木糖(13.41%)、甘露糖(3.52%)及鼠李糖(1.95%)(气相色谱-质谱分析).研究结果表明,桃胶中富含的多糖成分可能通过减轻DNA损伤机制,有效缓解重金属对皮肤细胞的毒性作用.
A heavy metal-induced human skin fibroblast(HFF-1)damage model was established using nickel sulfate(NiSO4).The protective effects of peach gum against heavy metal-induced cytotoxicity were evaluated through cell viability assays,morphological observation,and DNA damage marker(γ-H2AX)detection.Results demonstrated that exposure to NiSO4(8 mmol/L)for 12 h reduced HFF-1 cell viability to 44.65%(p<0.01)and significantly increased γ-H2AX expression in cells by 27.16-fold compared with the control.Pre-treatment with 8 mg/mL peach gum significantly improved cell viability to 59.22%(p<0.01)and decreased γ-H2AX expression to 3.8-fold of the control,indicating substantial protection against DNA damage.Component analysis revealed that the total sugar content of peach gum was 43.8%(phenol-sulfuric acid method).The polysaccharide fraction had an average molecular weight of 803.81 kDa(determined by high-performance liquid gel permeation chromatography)and was primarily composed of arabinose(47.96%),galactose(33.16%),xylose(13.41%),mannose(3.52%),and rhamnose(1.95%)as determined by gas chromatography-mass spectrometry(GC-MS).These findings suggest that the polysaccharide components in peach gum may alleviate heavy metal-induced cytotoxicity in skin fibroblasts,potentially through the mitigation of DNA damage mechanisms.
