富含原花青素的红山茶花提取液制备及其功效研究OA
Preparation and efficacy evaluation of a proanthocyanidin-rich Camellia sinensis extract
以浙江红山茶花为原料,优化大孔树脂D101对红山茶花中的原花青素纯化工艺,制备富含原花青素的红山茶花提取液并研究其功效活性.结果表明,大孔树脂D101纯化后红山茶花提取物中原花青素纯度达40.69%,较纯化前提高9.64倍.体外研究表明富含原花青素的红山茶花提取液显著清除1,1-二苯基-2-三硝基苯肼自由基(IC50=0.19%);红山茶花提取液含量为5%,对晚期糖化终产物的抑制率为47.81%±1.59%;红山茶花提取液含量为0.05%,可以显著促进内人永生化角质形成细胞愈合,愈合率为63.99%±3.89%.此外,其在含量为0.30%时,可以通过TGF-β1/Samds信号通路促进人正常成纤维细胞中Ⅰ型胶原蛋白和Ⅲ型胶原蛋白相对含量显著升高71.75%±3.55%和27.80%±1.49%,同时MMP-1含量显著降低63.38%±2.80%.
Using Zhejiang red camellia(Camellia sinensis)as the raw material,the purification process for proanthocyanidins was optimized using D101 macroporous resin.A proanthocyanidin-rich red camellia extract was prepared,and its functional activities were systematically evaluated.The results showed that purification with D101 macroporous resin increased the proanthocyanidin purity to 40.69%,representing a 9.64-fold enhancement compared with the unpurified extract.In vitro activity assays demonstrated that the proanthocyanidin-rich Camellia sinensis extract exhibited strong antioxidant capacity,effectively scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH)radicals with an IC50 value of 0.19%.At a concentration of 5%,the extract inhibited the formation of advanced glycation end products(AGEs)by 47.81%±1.59%.At 0.05%,the extract significantly promoted wound healing in human immortalized keratinocytes,achieving a healing rate of 63.99%±3.89%.Furthermore,treatment with 0.30%extract markedly increased the relative levels of type Ⅰ and type Ⅲ collagen in human normal fibroblasts by 71.75%±3.55%and 27.80%±1.49%,respectively,while reducing matrix metalloproteinase-1(MMP-1)expression by 63.38%±2.80%.These effects were mediated through activation of the TGF-β1/Smads signaling pathway.In conclusion,the proanthocyanidin-rich Camellia sinensis extract exhibits potent antioxidant,anti-glycation,skin repair,and anti-wrinkle activities,highlighting its potential application in functional cosmetics and skin care formulations.
