鱼酱排毒合剂调控HIF-1信号通路改善慢性鼻窦炎氧化应激的作用机制OA
Study on the mechanism of YuJiang Paidu Decoction(鱼酱排毒合剂)in regulating the HIF-1 signaling pathway to ameliorate oxidative stress in chronic rhinosinusitis
目的 探究鱼酱排毒合剂(YJPDHJ)调控 HIF-1 信号通路改善慢性鼻窦炎(CRS)氧化应激的作用机制.方法 OVA+AP滴鼻建立CRS小鼠模型,分为对照组(NC)、模型组(MC)、阳性药地塞米松组(Dexa)、鱼酱排毒合剂低剂量组(YJ-L)、高剂量组(YJ-H);0.4 mmol/L 叔丁基过氧化氢(TBHP)处理人鼻粘膜上皮细胞(HNEpCs)构建细胞模型.CCK-8 法检测细胞活力、HE 染色观察小鼠鼻腔,蛋白质印迹、qPCR 检测炎症因子、HIF-1 信号通路相关蛋白及基因表达,测定氧化应激指标.结果 与 MC 组比较,Dexa 组、YJ-H 和YJ-L组小鼠鼻腔病理结构改善,炎性细胞浸润减少,炎症因子 IL-4、IL-5、IL-13 表达受抑制,氧化应激状态缓解,且 YJ-H 组效果优于 YJ-L 组.鱼酱排毒合剂呈剂量依赖性降低细胞内 ROS、GSSH/GSH 和MDA 水平,增强 SOD 和 CAT 活性,抑制 HIF-1α、VEGF 和 VEGF2 蛋白表达.结论 鱼酱排毒合剂通过调节 HIF-1 信号通路缓解 CRS 的炎症反应和氧化应激,抑制疾病病理发展.
Objective To investigate the mechanism of YuJiang Paidu Decoction(鱼酱排毒合剂,YJPDD)in ameliorating oxidative stress in chronic rhinosinusitis(CRS)by regulating the HIF-1 signaling pathway.Methods A CRS mouse model was established using OVA+AP nasal drops.Mice were divided into a control group(NC),a model control(MC),a positive control dexamethasone group(Dexa),a low-dose YJPDD group(YJ-L),and a high-dose YJPDD group(YJ-H).Human nasal epithelial cells(HNEpCs)were treated with 0.4 mmol/L tert-butyl hydroperoxide(TBHP)to to construct a cell model.Cell viability was assessed via CCK-8 assay,while mouse nasal cavities was observed using hematoxylin-eosin(HE)staining.Western blot(WB)and qPCR were performed to evaluate inflammatory factors,HIF-1 pathway pathway-related proteins and gene expression,and oxidative stress indicators were assessed.Results Compared with the MC group,the Dexa,YJ-H,and YJ-L groups showed improved pathological structure of the nasal cavity,reduced inflammatory cell infiltration,and suppressed expression of inflammatory factors IL-4,IL-5,and IL-13,and alleviated oxidative stress,with the superior effect of the YJ-H group compare to the YJ-L group.YJPDD demonstrating dose-dependent reduc-tions in intracellular ROS,GSSG/GSH,and MDA levels,alongside enhanced SOD and CAT activity,and inhibited the protein expres-sion of HIF-1α,VEGF,and VEGFR2.Conclusion YJPDD ameliorates inflammatory response and oxidative stress in CRS by regulat-ing the HIF-1 signaling pathway,thereby inhibiting the pathological progression of the disease.
