新风胶囊调节PI3K/AKT信号通路减轻骨关节炎大鼠炎症及骨代谢紊乱的机制研究OA
Mechanism of Xinfeng Capsule(新风胶囊)in regulating the PI3K/AKT signaling pathway to alleviate inflammation and bone metabolism disorder in osteoarthritis rats
目的 探究新风胶囊(XFC)通过PI3K/AKT信号通路对骨关节炎(OA)大鼠炎症及骨代谢的影响.方法 64只SD大鼠等分为8组:对照(Control)组,模型(Model)组,XFC低、中、高剂量(XFC-L、XFC-M、XFC-H)组,塞来昔布(Celecoxib)组,SC79(AKT激动剂)组,XFC-H+SC79组;对除Control组外大鼠以胶原蛋白酶制备OA模型.番红固绿、阿利辛蓝染色进行软骨组织学分析;ELISA检测血清中TGF-β1、TNF-α、IL-1β、IL-6、TRACP-5b、IGF-1、OPG、BALP表达;免疫印迹检测软骨中PI3K、p-PI3K、AKT、p-AKT 蛋白表达.结果 与Control组比较,Model组软骨表面粗糙,软骨细胞及细胞外基质减少,潮线破坏;IL-1β、IL-6、TNF-α、TGF-β1、TRACP-5b、p-PI3K、p-AKT表达升高(P<0.05),OPG、BALP、IGF-1 表达降低(P<0.05).与Model组比较,XFC可以不同程度降低IL-1β、IL-6、TNF-α、TGF-β1、TRACP-5b、p-PI3K、p-AKT表达(P<0.05),升高OPG、BALP、IGF-1表达(P<0.05),减轻软骨损伤;SC79组显示出更严重的炎症、骨代谢紊乱及软骨损伤.与XFC-H组比较,SC79可以部分逆转XFC-H对炎症、骨代谢紊乱及软骨损伤的改善作用.结论 XFC调节PI3K/AKT信号通路减轻OA大鼠炎症及骨代谢紊乱,改善软骨损伤.
Objective To investigate the effects of Xinfeng Capsule(新风胶囊,XFC)on inflammation and bone metabolism in osteoar-thritis(OA)rats via the PI3K/AKT signaling pathway.Methods Sixty-four SD rats were randomly divided into eight groups:the Control group,Model group,low/medium/high-dose XFC(XFC-L,XFC-M,XFC-H)groups,celecoxib group,AKT activator SC79 group,and XFC-H+SC79 group.The OA Model was induced in all groups except the Control group by intra-articular injection of collagenase.Cartilage histology was analyzed using Safranin O/Fast Green and Alcian blue staining.Serum levels of TGF-β1,TNF-α,IL-1β,IL-6,TRACP-5b,IGF-1,OPG,and BALP were measured by ELISA.The protein expression levels of PI3K,p-PI3K,AKT,and p-AKT in cartilage were detected by Western blot(WB)analysis.Results Compared with the Control group,the Model group presented with a rough cartilaginous surface,reduced chondrocytes and extracellular matrix,and a destroyed tidemark.The expression levels of IL-1β,IL-6,TNF-α,TGF-β1,TRACP-5b,p-PI3K,and p-AKT were significantly elevated(P<0.05),whereas the expression levels of OPG,BALP,and IGF-1 were significantly reduced(P<0.05).Compared with the Model group,XFC treatment resulted in reduced expression levels of IL-1β,IL-6,TNF-α,TGF-β1,TRACP-5b,p-PI3K,and p-AKT(P<0.05),increased expression levels of OPG,BALP,and IGF-1(P<0.05),and alleviated cartilage damage.The SC79 group exhibited more severe inflammation,bone metabolism disorders,and cartilage damage compared with the Model group.Compared with the XFC-H group,co-administration of SC79 partially reversed the ameliorative effects of XFC-H on inflammation,bone metabolism disorder,and cartilage damage.Conclu-sion XFC alleviates inflammation and bone metabolism disorders in OA rats and ameliorates cartilage damage by regulating the PI3K/AKT signaling pathway.
