孟河医派特色炮制辅料猪心血和猪血冻干粉质量标准研究OA
Quality standard research on freeze-dried powders of porcine cardiac blood and porcine blood as characteristic processing excipients of Menghe medical school
目的 建立孟河医派炮制辅料猪心血(PCB)和猪血(PB)的质量标准.方法 收集15批次的猪心血和猪血,对其进行性状、聚合酶链式反应(PCR)、红外光谱和肽指纹图谱结合化学计量学鉴别以及水分、总灰分、酸不溶性灰分、浸出物、铁元素、氮元素、转铁蛋白(Tf)和水解氨基酸含量测定.结果 基于所建立的方法对样品进行系统测定,结果显示,可通过PCR反应鉴别PCB、PB和其他动物血,可通过红外光谱和肽指纹图谱结合化学计量学方法区分PCB和PB,建议PCB和PB的水分均不得超过8.0%,PCB总灰分不得超过8.0%,PB总灰分不得超过11.0%,PCB酸不溶性灰分不得超过0.50%,PB酸不溶性灰分不得超过1.20%,浸出物含量均不得少于42.0%,含氮量均不得少于10.0%,Tf含量均不得少于0.10%,PCB中铁元素含量不得少于0.20%,PB中铁元素含量不得少于0.18%,PCB中水解氨基酸含量不得少于36.0%,PB中水解氨基酸含量不得少于32.0%.结论 该研究补充并完善了辅料猪心血和猪血的质量评价标准,为制定猪心血、猪血及其炮制饮片质量标准提供科学依据.
Objective To establish the quality standard for porcine cardiac blood(PCB)and porcine blood(PB)as processing excipi-ents of Menghe Medical School.Methods Fifteen batches of PCB and PB samples(as freeze-dried powders)were collected.They were identified by traits,polymerase chain reaction(PCR),infrared spectroscopy,and peptide fingerprinting combined with chemometrics.The contents of moisture,total ash,acid-insoluble ash,extract,iron,nitrogen,transferrin(Tf),and hydrolyzed amino acids were determined.Results The samples were systematically analyzed using the established methods.The results showed that PCB,PB,and other animal blood could be differentiated by PCR.PCB and PB could be distinguished by infrared spectroscopy and peptide fingerprint-ing combined with chemometrics.It was recommended that the moisture content in both PCB and PB should not exceed 8.0%;the total ash in PCB should not exceed 8.0%,and that in PB should not exceed 11.0%;the acid-insoluble ash in PCB should not exceed 0.50%,and that in PB should not exceed 1.20%;the extract content should not be less than 42.0%;the nitrogen content should not be less than 10.0%;the Tf content should not be less than 0.10%;the iron content in PCB should not be less than 0.20%,and that in PB should not be less than 0.18%;the hydrolyzed amino acids content in PCB should not be less than 36.0%,and that in PB should not be less than 32.0%.Conclusion This study supplements and improves the quality evaluation criteria for PCB and PB as excipients,provid-ing a scientific basis for establishing quality standards for PCB,PB,and their processed decoction pieces.
