补肾调肝中药调控线粒体自噬治疗帕金森病进展OA
Progress in kidney‑tonifying and liver‑regulating traditional Chinese medicines for Parkinson's disease via regulating mitophagy
线粒体自噬异常与帕金森病(PD)肝肾亏虚的中医病机联系密切,是该病发生发展的关键环节.综述补肾调肝中药通过调控线粒体自噬治疗PD的研究进展,补肾调肝中药有效成分及复方制剂在调控线粒体自噬治疗PD运动或非运动症状功效方面存在一致性,其作用核心大多围绕[丝氨酸/苏氨酸蛋白激酶1(PINK1)]/E3泛素连接酶(Parkin)这一关键通路,通过促进或抑制线粒体自噬发挥神经保护作用,也可通过受体介导、沉默信息调节因子1(SIRT1)/叉头框蛋白O(FoxO)通路等调控线粒体自噬.以线粒体自噬为靶点,有望阐明补肾调肝中药治疗PD的现代生物学内涵.
Abnormal mitophagy is closely associated with the traditional Chinese medicine(TCM)pathogenesis of"liver-kidney deficiency"in Parkinson's disease(PD)and serves as a key link in the onset and progression of the disease.This review summarizes the research progress of kidney-tonifying and liver-regulating TCMs in treating PD by regulating mitophagy.Both active components and compound preparations of kidney-tonifying and liver-regulating TCMs show consistent efficacy in improving motor and non-motor symptoms of PD through mitophagy regulation.Their core mechanisms primarily focus on the serine/threonine-protein kinase 1(PINK1)/E3 ubiquitin ligase(Parkin)pathway,exerting neuroprotective effects by promoting or inhibiting mitophagy.Additionally,they can regulate mitophagy via receptor-mediated pathways or the sirtuin 1(SIRT1)/forkhead box O(FoxO)pathway.Targeting mitophagy is expected to clarify the modern biological connotations of kidney-tonifying and liver-regulating TCMs in PD treatment.
