目的 建立加味桔梗汤多成分的超高效液相色谱(UPLC)含量测定方法,并应用Box-Behnken 响应面法对其水提醇沉工艺进行优化.方法 采用UPLC串联蒸发光散射检测器(UPLC-ELSD)法测定桔梗皂苷D的质量分数,UPLC串联紫外检测器(UPLC-UV)法测定没食子酸、绿原酸、柯里拉京、橙皮苷、迷迭香酸的含量;基于层次分析法(AHP)-熵权法确定各指标的权重系数;在单因素考察的基础上,采用Box-Behnken响应面法考察浸泡时间、煎煮时间对水提工艺的影响,并考察乙醇体积分数、醇沉时间、药液密度对醇沉工艺的影响.结果 建立了加味桔梗汤多成分的UPLC含量测定方法,并优选出其最佳水提工艺为不浸泡、加水12倍、煎煮65 min、煎煮2次,最佳醇沉工艺为乙醇体积分数80%、醇沉48 h、药液浓度1.65 g·mL-1.结论 建立的含量测定方法准确可靠,筛选得到的工艺参数稳定,可为加味桔梗汤的后续研究开发提供依据.

Objective To establish a validated ultra-performance liquid chromatography(UPLC)method for the simultaneous quantification of multiple components in modified Jiegeng Decoction and to optimize its water extraction and ethanol precipitation processes using the Box-Behnken design-response surface method(BBD-RSM).Methods Platycodin D was determined by UPLC coupled with evaporative light scattering detection(UPLC-ELSD),while gallic acid,chlorogenic acid,corilagin,hesperidin,and rosmarinic acid were quantified via UPLC with ultraviolet detection(UPLC-UV).Weight coefficients for each evaluation index were calculated using the analytic hierarchy process(AHP)-entropy weight method.Based on the results of single-factor experiments,BBD-RSM was employed to investigate the effects of soaking time and decoction time on the water extraction process,as well as the impacts of ethanol concentration,precipitation time,and extract density on the ethanol precipitation process.Results A UPLC method for the simultaneous determination of multiple components in modified Jiegeng Decoction was successfully established and validated.The optimized water extraction conditions were confirmed as follows:no soaking,12-fold volume of water relative to the raw material,and two rounds of decoction(65 minutes per round).For ethanol precipitation,the optimal parameters were determined to be 80%(V/V)ethanol concentration,48 hours of precipitation,and an extract density of 1.65 g·mL-1.Conclusions The established UPLC method exhibits high accuracy and reliability,and the optimized extraction-precipitation parameters demonstrate good stability.This study provides a solid technical foundation for subsequent pharmacological research and large-scale industrial production of Modified Jiegeng Decoction.
