基于SDF-1/CXCR4轴探讨加味小青龙汤对寒性哮喘大鼠BMSCs归巢的影响OA
Effect of Jiawei Xiaoqinglong Decoction(加味小青龙汤)on BMSCs homing in rats with cold asthma via the SDF-1/CXCR4 axis
目的 探讨加味小青龙汤经基质细胞衍生因子-1(SDF-1)/趋化因子受体4(CXCR4)轴对骨髓间充质干细胞(BMSCs)归巢效率的影响,分析其联合BMSCs移植治疗寒性哮喘的作用机制.方法 将50只雄性SD大鼠随机分为空白组、模型组、BMSCs模型组、BMSCs模型+加味小青龙汤组、BMSCs模型+地塞米松组.OVA联合氢氧化铝致敏、雾化激发及空调冷风吹袭建立寒性哮喘模型.BMSCs模型组尾静脉注射CFSE-BMSCs悬液1 mL(1×106/mL),在此基础上用药组给予加味小青龙汤水煎剂(6.39 g/kg)或地塞米松混悬液(0.125 mg/kg)灌胃.7天后评估大鼠哮喘症状,HE染色观察肺组织病理学变化,免疫荧光观察BMSCs分布,ELISA检测BALF中IL-6、IL-10水平,Real-time PCR和Western blot检测肺组织中SDF-1/CXCR4 mRNA和蛋白表达水平.结果 模型组大鼠哮喘症状及肺组织炎症评分升高,BALF中IL-6升高而IL-10降低(P<0.01),各治疗组上述指标均显著改善(P<0.05或P<0.01).BMSCs模型+加味小青龙汤组可见归巢BMSCs数量显著多于其它移植组,且肺组织中SDF-1、CXCR4 mRNA和蛋白表达水平最高(P<0.05或P<0.01).结论 加味小青龙汤可通过上调SDF-1/CXCR4信号轴,提高BMSCs向寒性哮喘肺组织归巢效率,进而增强抗炎和哮喘症状改善功效.
Objective To investigate the effect of Jiawei Xiaoqinglong Decoction(加味小青龙汤,JXD)on bone marrow mesenchymal stem cells(BMSCs)homing efficiency through the stromal cell-derived factor-1(SDF-1)/C-X-C chemokine receptor type 4(CXCR4)axis,and to analyze its therapeutic mechanism in combination with BMSCs transplantation for cold asthma.Methods Fifty male Sprague-Dawley(SD)rats were randomly divided into five groups:control,model,BMSCs transplantation,BMSCs transplantation+JXD,and BMSCs transplantation+dexamethasone.A cold asthma model was established using ovalbumin(OVA)and aluminum hydroxide sensitization,nebulization challenge,and cold air exposure.Rats in the transplantation groups received 1 mL of CFSE-labeled BMSCs suspension(1×10⁶ cells/mL)via tail vein injection.The treatment groups were subsequently administered JXD water extract(6.39 g/kg)or dexamethasone suspension(0.125 mg/kg)by gavage for 7 days.Asthma symptoms were evaluated,and lung pathology was examined by hematoxylin-eosin(HE)staining.BMSCs distribution was observed by immunofluorescence(IF),while interleukin-6(IL-6)and interleukin-10(IL-10)levels in bronchoalveolar lavage fluid(BALF)were measured by enzyme-linked immu-nosorbent assay(ELISA).The mRNA and protein expression of SDF-1 and CXCR4 in the lung tissues was detected by quantitative real-time PCR(qRT-PCR)and Western blot(WB)analysis,respectively.Results The model group showed aggravated asthma symptoms,increased lung inflammation scores,elevated IL-6 levels,and decreased IL-10 levels in BALF(all P<0.01).All treatment groups demonstrated significant improvements in these indicators(P<0.05 or P<0.01).The BMSCs+JXD group exhibited the highest number of homed BMSCs and the most pronounced upregulation of SDF-1 and CXCR4 mRNA and protein expression(P<0.05 or P<0.01).Conclusion JXD may enhance BMSCs homing to lung tissues in cold asthma by upregulating the SDF-1/CXCR4 signaling axis,thereby amplifying its anti-inflammatory and therapeutic effects.
