目的 探讨补肾健脾方通过调控HIF-1α/BNIP3通路介导的线粒体自噬,影响细胞焦亡,从而改善骨质疏松(OP)大鼠骨状态的分子机制.方法 将60只SD大鼠随机分为空白组、模型组、福善美组、补肾健脾方高、中、低剂量组,除空白组外均采用地塞米松建立OP模型,模型建立同时予以药物干预,连续5周,结束后进行相应指标测定.酶联免疫检测法检测大鼠ALP、PINP、IL-1β、IL-18的含量;Micro-CT进行各组大鼠股骨远端三维图像重建并检测骨密度;HE染色进行各组大鼠股骨组织的病理观察;蛋白免疫印迹法检测大鼠股骨组织中线粒体自噬蛋白HIF-1α、BNIP3、p62、LC3蛋白表达情况;检测细胞焦亡关键蛋白NLRP3、Caspase-1、GSDMD表达情况.结果 补肾健脾方能够提高骨密度,改善骨组织结构;上调HIF-1α和BNIP3的表达,促进线粒体自噬相关蛋白LC3-II/LC3-I比值及p62的表达,下调IL-1β、IL-18、ROS表达,抑制NLRP3、Caspase-1和GSDMD的表达.结论 补肾健脾方通过调控HIF-1α/BNIP3通路介导的线粒体自噬,抑制细胞焦亡,从而改善骨质疏松大鼠的骨状态.

Objective To investigate the molecular mechanism through which Bushen Jianpi Formula(补肾健脾方,BSJPF)amelio-rates bone status in osteoporotic rats,by regulating the HIF-1α/BNIP3 pathway-mediated mitochondrial mitophagy and influencing pyroptosis.Methods Sixty SD rats were randomly assigned to six groups:the blank,model,alendronate,and high/medium/low-dose BSJPF groups.Except for the blank group,an osteoporosis(OP)model was induced in all rats using dexamethasone.Drug interven-tions were administered concurrently for 5 consecutive weeks.Relevant indicators were measured after the intervention.Serum levels of ALP,PINP,IL-1β,and IL-18 were measured using ELISA.Micro-CT was performed for 3D reconstruction and bone mineral density(BMD)analysis of the distal femur.Histopathological changes in femoral tissues were observed via HE staining.The protein expression levels of mitophagy-related markers(HIF-1α,BNIP3,p62,and LC3)and pyroptosis-related markers(NLRP3,Caspase-1,and GSDMD)in femoral tissues were detected by Western blot(WB).Results BSJPF treatment significantly increased bone mineral density and ameliorated the histopathological structure of bone tissue.It upregulated the expression of HIF-1α and BNIP3,increased the LC3-II/LC3-I ratio,and modulated p62 expression.Concurrently,BSJPF downregulated the serum levels of IL-1β and IL-18,reduced reac-tive oxygen species(ROS)levels,and inhibited the expression of NLRP3,Caspase-1,and GSDMD proteins.Conclusion BSJPF allevi-ates bone deterioration in osteoporotic rats by regulating HIF-1α/BNIP3-mediated mitophagy and inhibiting pyroptosis.
