橡胶树三倍体品种云研73-46花药愈伤组织诱导及体胚发生技术研究OA
Anther Callus Induction and Somatic Embryogenesis inTtriploid Rubber Tree Variety Yunyan 73-46
为建立橡胶树三倍体品种云研 73-46 花药体胚植株再生技术体系,本研究以其花药为外植体,采用正交试验设计探讨不同激素组合对愈伤组织诱导及体胚发生效率的影响,并比较不同培养时长下愈伤组织的形态特征与体胚发生能力.结果表明:(1)激素对愈伤组织诱导至关重要,无激素条件下花药无法脱分化产生愈伤并于 10 d后褐化死亡,而在含有 2,4-D、NAA、KT及TDZ的培养基中,花药可脱分化产生愈伤组织,诱导率的变幅为 15%~91.56%,均值为(69.41±30.37)%.4 种激素对愈伤组织诱导率及后续体胚发生率的影响差异极显著(P<0.01),影响顺序为 2,4-D>TDZ>NAA>KT.综合筛选得到最优激素组合为 2,4-D 1.0 mg/L+NAA 0.8 mg/L+TDZ 0.02 mg/L.经过验证,其愈伤组织诱导率和后续体胚发生率分别为(86.15±1.14)%和(14.46±0.89)%,相较于最佳试验组合(G7)分别显著提高 2.31%和 5.40%;(2)该品种体胚发生能力普遍较弱,体胚发生率的变幅为 6.91%~19.31%,均值为(10.58±3.95)%.6-BA、ABA、NAA及KT对体胚发生率的影响差异极显著(P<0.01),影响顺序为ABA>6-BA>NAA>KT,最优组合为 6-BA 0.4 mg/L+ABA 0.2 mg/L+NAA 0.2 mg/L+KT 0.2 mg/L(M9),体胚发生率达(19.31±0.60)%,显著高于其他处理;(3)55 d是愈伤组织适宜培养时长,愈伤生长状态较佳,无褐化,且体胚发生能力最强,体胚发生率为(30.83±1.67)%,子叶形体胚发生率为(18.33±0.83)%,平均体胚数为(0.63±0.03)个,极显著高于其他培养时长.通过优化云研 73-46 花药愈伤组织诱导以及体胚发生阶段的激素配比,同时调整愈伤诱导培养时长,能够显著提升该品种花药体胚植株的培育效率,为构建橡胶树三倍体品种体胚植株规模化繁育体系提供技术支撑.
To establish a regeneration system for somatic embryo-derived plants from the anthers of the triploid rubber tree clone Yunyan 73-46,this study utilized anthers as explants and employed an orthogonal experimental design to examine the effects of various hormone combinations on callus and somatic embryo induction efficiency.Furthermore,the morphological characteristics and differentiation capacity of callus under different culture duration were analyzed.Hormones were crucial for callus induction.In the absence of hormones,anthers failed to dedifferentiate and underwent browning and necrosis after 10d.In media containing 2,4-D,NAA,KT and TDZ,anther dedifferentiation to produce callus occurred with induction rates ranging from 15%to 91.56%,with a mean of(69.41±30.37)%.The impact of 2,4-D,NAA,KT and TDZ on callus induction rate and subsequent somatic embryogenesis rate were all highly significant(P<0.01),with the order of influence being 2,4-D>TDZ>NAA>KT.The optimal combination identified through com-prehensive screening was 2,4-D 1.0 mg/L+NAA 0.8 mg/L+TDZ 0.02 mg/L.Upon verification,the callus induction rate and subsequent somatic embryogenesis rate was(86.15±1.53)%and(14.46±0.89)%,respectively,marking significant improvements of 2.31%and 5.40%over the best experimental combination(G7).The somatic embryogenesis capacity of this clone was generally low,with somatic embryo formation rates ranging from 6.91%to 19.31%,with a mean of(10.58±3.95)%.The effects of 6-BA,ABA,NAA and KT on somatic embryogenesis rate were highly significant(P<0.01),with the order of influence being ABA>6-BA>NAA>KT.The optimal combination was 6-BA 0.4 mg/L+ABA 0.2 mg/L+NAA 0.2 mg/L+KT 0.2 mg/L(M9),achieving a somatic embryogenesis rate of(19.31±0.60)%,which was significantly higher than other treatments.The optimal culture duration for callus was determined to be 55 d,which exhibited superior growth status without browning and the strongest somatic embryo differentiation capacity.The so-matic embryogenesis rate was(30.83±1.67)%,the cotyledonary embryo formation rate was(18.33±0.83)%,and the av-erage number of somatic embryos was(0.63±0.03),all of which were extremely significantly higher than those at other culture durations.By optimizing the hormone ratios during callus induction and somatic embryogenesis stages of Yu-nyan 73-46 anthers,coupled with adjusting the duration of callus induction culture,the production efficiency of an-ther-derived somatic embryo plantlets can be significantly enhanced.This approach would provide technical support for establishing a large-scale propagation system of somatic embryo plantlets for triploid rubber tree clones.
