内异方对子宫内膜异位症大鼠异位子宫内膜细胞凋亡的影响OA
Effects of Neiyi Prescription on Ectopic Endometrial Cell Apoptosis in Endometriosis Rats
目的 探究内异方(Neiyi Prescription,NYP)对异位子宫内膜组织细胞凋亡的影响及相关机制.方法 将24只SD雌性大鼠按照随机数字表法分为对照组(6只,生理盐水2 ml/d)、模型组(6只,生理盐水2 ml/d)、NYP低剂量组(6只,0.63 g/kg)、NYP高剂量组(6只,1.26 g/kg),除对照组外均采用自体移植法构建子宫内膜异位症(Endometriosis,EMs)大鼠模型.每组大鼠均接受相应干预3周,灌胃2次/d.计算NYP对异位子宫内膜囊肿的抑制率;苏木精-伊红(Hematoxylin-eosin,HE)染色法观察大鼠异位子宫内膜组织形态;末端脱氧核苷酸转移酶dUTP缺口末端标记(Terminal deoxynucleotidyl transferase dUTP nick end labeling,TUNEL)染色法检测细胞凋亡率;免疫组织化学法(Immunohistochemistry,IHC)检测蛋白激酶B(Protein kinase B,AKT1)的表达.结果 与对照组比较,模型组上皮细胞排列紧密、不规则,间质细胞增多;细胞凋亡率减少(P<0.001),AKT1阳性率增加(P<0.001).模型组大鼠经生理盐水干预后,囊肿体积较干预前增大;经NYP低、高剂量治疗后,囊肿体积较治疗前均有不同程度缩小.与模型组比较,NYP低、高剂量组对异位子宫内膜囊肿的抑制率差异有统计学意义(P<0.05);NYP低剂量组的上皮细胞排列整齐,间质细胞减少;与模型组比较,NYP低、高剂量组的细胞凋亡率呈剂量依赖性升高(P<0.001);NYP组的AKT1阳性率减少(P<0.01).与NYP低剂量组比较,NYP高剂量组的上皮细胞排列疏松,间质细胞减少,分布更均匀.结论 高剂量NYP可能通过下调EMs大鼠异位子宫内膜囊肿中AKT1的表达,促进其细胞凋亡,从而抑制囊肿的生长,发挥治疗作用.
Objective To investigate the effects and possible mechanisms of Neiyi Prescription(NYP)on ectopic endometrial tissue apoptosis.Methods Twenty-four female SD rats were randomly assigned to a control group(2 ml/d of normal saline,n=6),a model group(2 ml/d of normal saline,n=6),a low-dose NYP group(0.63 g/kg,n=6),and a high-dose NYP group(1.26 g/kg,n=6).Except for the control group,the other groups received autologous transplantation to construct an endometriosis(EMs)rat model.Gavage was performed on every group twice a day for 3 consecutive weeks.Afterwards,the following parameters were determined by different methods:the inhibitory rate on ectopic endometrial cysts by NYP calculated using formula,tissue morphology by the hematoxylin-eosin(HE)staining,apoptosis rate by the terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL)staining,and Protein Kinase B(AKT1)expression by the immunohistochemical(IHC)staining.Results Compared with the control group,the model group exhibited tightly packed epithelial cells in an irregular manner,increased interstitial cells,decreased apoptosis rate(P<0.001),and increased AKT1 positivity(P<0.001).After intervention with normal saline,the model group exhibited increased cyst volume,while the low-and high-dose NYP groups exhibited decreased cyst volume to varying degrees.Compared to the model group,a significant inhibitory rate on ectopic endometrial cysts was observed in both low-and high-dose NYP groups(P<0.05).In the low-dose NYP group,the epithelial cells were more regularly arranged,with a decreased number of interstitial cells.In both low-and high-dose NYP groups,a dose-dependent increase was observed in apoptosis rate(P<0.001).The AKT1 positivity rate was reduced in the NYP groups(P<0.01).Compared to the low-dose NYP group,the high-dose NYP group exhibited looser epithelial cell arrangement and decreased interstitial cells in a more uniform distribution.Conclusion High-dose NYP may exert therapeutic effects by down-regulating AKT1 expression,thereby promoting apoptosis and inhibiting ectopic endometrial cyst growth in EMs rats.
