Notch通路基因Djaph-1、Djcsl与Djnumb在涡虫再生中的功能研究OA
Functional roles of the Notch pathway genes Djaph-1,Djcsl and Djnumb in planarian regeneration
Notch信号通路是一种保守的细胞间通讯机制,在组织发育与稳态维持中发挥重要作用.为探究其在组织再生及干细胞调控中的作用,本研究选择涡虫为实验材料.鉴定了日本三角涡虫中的Notch信号通路基因aph-1、csl、numb的同源基因,并探讨了它们在涡虫再生过程中的生物学功能.研究结果显示,Djaph-1、Djcsl与Djnumb mRNA在整体涡虫中广泛表达,并在再生涡虫的芽基中富集表达.Djaph-1、Djcsl与Djnumb干扰均导致再生涡虫出现身体中线黑斑和眼点不对称的异常表型以及中枢神经系统再生缺陷,引起了不同再生时间点细胞增殖水平的变化以及眼点、表皮相关基因的表达变化.进一步研究表明,Djaph-1和Djcsl均可正调控Notch信号,而Djnumb则发挥负向调控作用.综上,Djaph-1、Djcsl与Djnumb参与调控干细胞增殖与分化,在涡虫再生过程中发挥关键作用.
The Notch signaling pathway is a conserved intercellular communication mechanism that plays a crucial role in tissue development and homeostasis.To investigate its functions in tissue regeneration and stem cell regulation,planarians were used as an experimental model.Homologous of the Notch signaling pathway genes aph-1,csl,and numb were identified in Dugesia japonica and their biological functions during planarian regeneration were characterized.The results showed that Djaph-1,Djcsl,and Djnumb mRNA were broadly expressed in whole planarians and were highly enriched in the regenerating blastema.Knock-down of Djaph-1,Djcsl,and Djnumb led to abnormal phenotypes such as black spots along the body axis and asymmetric eye spots,as well as defects in central nervous system regeneration.These phenotypes were ac-companied by altered cell proliferation dynamics at different regeneration stages and chenges in the expression of eye-and epidermis-related genes.Further studies demonstrated that Djaph-1 and Djcsl positively regulate Notch signaling,whereas Djnumb functions as a negative regulator.Collectively,these results suggest that Djaph-1,Djcsl,and Djnumb are key regulators of stem cell proliferation and differentiation and play crucial roles in planarian regeneration.
