基于网络药理学与分子对接技术探究当归四逆汤治疗糖尿病足溃疡的作用机制OA
Exploring the mechanism of Danggui Sini Decoction in treating diabetic foot ulcer based on network pharmacology and molecular docking technology
目的:基于网络药理学方法与分子对接技术探究当归四逆汤治疗糖尿病足溃疡(DFU)的作用机制.方法:通过TCMSP数据库检索当归四逆汤的活性成分以及作用靶点,通过GeneCards、OMIM数据库检索DFU疾病靶点,利用Cyto-scape 3.7.1软件构建"中药-活性成分-靶点"网络图和蛋白质-蛋白质互作(PPI)网络图;利用DAVID数据库进行基因本体(GO)功能及京都基因与基因组百科全书(KEGG)通路富集分析,并通过微生信平台将结果可视化;通过CB-Dock2平台将关键靶点与活性成分进行分子对接.结果:共收集到141个当归四逆汤活性成分及247个相关靶点,DFU疾病靶点2255个,药物与疾病交集靶点149个,其中度值(degree)排名前5位的靶点分别是丝氨酸-苏氨酸蛋白激酶(Akt1)、白蛋白(ALB)、肿瘤坏死因子(TNF)、白细胞介素-6(IL-6)、白细胞介素-1β(IL-1β).GO功能与KEGG通路富集分析表明,当归四逆汤有效成分可能通过作用于晚期糖基化终末产物/晚期糖基化终产物受体(AGE/RAGE)信号通路和肿瘤坏死因子(TNF)信号通路中的关键节点,抑制促炎信号传导,减轻氧化损伤,并改善局部血供,从而为溃疡愈合营造有利的微环境.分子对接分析表明,具有代表性的组分(槲皮素、豆甾醇、柚皮素、山奈酚、β-谷甾醇)与关键靶点紧密结合.结论:当归四逆汤通过多成分、多靶点和多途径的协同作用治疗DFU,加深了对当归四逆汤治疗DFU潜在分子机制的认识,为进一步的实验研究奠定了基础.
Objective:To investigate the mechanism of Danggui Sini Decoction(Angelica Cold Extremities Decoc-tion)in treating diabetic foot ulcer(DFU)using network pharmacology and molecular docking technology.Methods:Active components and targets of Danggui Sini Decoction were retrieved from the TCMSP database,while disease targets of DFU were obtained from the GeneCards and OMIM database.Cytoscape 3.7.1 software was used to construct a"drug-active component-target"network and a protein-protein interaction network.Gene Ontology(GO)functional enrichment and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analyses were performed using the DAVID database,with re-sults visualized via the microbiolink platform.Molecular docking of key targets and active components was conducted using the CB-Dock2 platform.Results:A total of 141 active components and 247 related targets of Danggui Sini Decoction were collected,along with 2255 disease targets for DFU.There were 149 overlapping targets,among which the top five targets by degree value were Akt1(protein kinase B1),ALB(albumin),TNF(tumor necrosis factor),IL-6(interleukin-6),and IL-1β(interleukin-1 beta).GO function and KEGG pathway enrichment analysis indicate that the active components of Danggui Sini Decoction may inhibit pro-inflammatory signaling,alleviate oxidative damage,and improve local blood supply by acting on key nodes in the advanced glycation end product/receptor for advanced glycation end product(AGE/RAGE)signaling pathway and tumor necrosis factor(TNF)signaling pathway,thereby creating a favorable microenvironment for ulcer healing.Molecular docking analysis reveals that representative components(quercetin,stigmasterol,naringenin,kaempferol,and β-sitosterol)bind tightly to key targets.Conclusion:Danggui Sini Decoction treats DFU through synergis-tic effects of multiple components,targets,and pathways.This study enhances the understanding of the potential molecular mechanisms of Danggui Sini Decoction in treating DFU and provides a foundation for further experimental research.
