大麻二酚可溶性微针的制备、表征及初步药效学研究OA
Preparation,characterization and preliminary pharmacodynamic evaluation of cannabidiol-loaded dissolving microneedles
目的 制备大麻二酚(CBD)可溶性微针,并对其进行性能表征及初步药效学评价,旨在实现药物精准递送,减少给药剂量,降低不良反应,从而优化疗效与安全性之间的平衡.方法 采用分层浇筑法制备 CBD 可溶性微针,通过单因素试验优化制备工艺,对微针的形貌、机械性能等进行质量评价,并采用小鼠醋酸扭体模型考察CBD可溶性微针的体内镇痛效果.结果 CBD可溶性微针的最优制备工艺为:分别以PVP K30 和PVP K90 为针尖和背衬材料,采用两步离心法制备,针尖溶液离心时间为 5 min,固定离心机转速为4500 r·min-1.所得微针形态完整,机械强度良好,单针承压力不低于 2.9 N.药效学结果显示,CBD可溶性微针在给药总量远低于凝胶组的情况下,仍表现出更高的疼痛抑制率.结论 本研究成功制备了CBD可溶性微针,其在极低剂量下仍表现出明显的镇痛效果,显示出良好的应用前景.
Objective To prepare cannabidiol(CBD)-loaded dissolving microneedles(DMNs),and characterize their properties and conduct a preliminary pharmacodynamic evaluation,aiming to achieve precise drug delivery,reduce dosage,minimize adverse effects,and thereby optimize the balance between efficacy and safety.Methods CBD-loaded DMNs were fabricated using a layer-by-layer casting method,with the preparation process optimized through single-factor experiments.The morphology and mechanical properties of the microneedles were evaluated for quality assessment.The in vivo analgesic efficacy was investigated using a mouse acetic acid-induced writhing model.Results The optimal preparation parameters were determined as follows:using PVP K30 and PVP K90 as the matrix materials for the needle tips and backing layer,re-spectively,employing a two-step centrifugation method with a tip solution centrifugation time of 5 min and a fixed centrifuge speed of 4 500 r·min-1.The resulting microneedles exhibited intact morphology and satisfactory mechanical strength,with a single needle fracture force of no less than 2.9 N.Pharmacodynamic results showed that the CBD-loaded DMNs exhibited a higher pain inhibition rate compared to the CBD gel formulations,despite being administered at a much lower dose.Conclu-sion This study successfully developed CBD-loaded dissolving microneedles,which demonstrated significant analgesic effi-cacy at an extremely low dosage,indicating promising application prospects.
