目的 通过网络药理学分析和体内实验验证,探讨胡芦巴对糖尿病肾病的抗炎及抗氧化作用.方法 利用TCMSP、SwissTarget和UniProt数据库筛选、补充和校正胡芦巴有效成分及相关作用靶点;使用GeneCard、DisG-Net、DigSee、TTD、OMIM和DrugBank数据库获取糖尿病肾病的相关靶点;使用STRING 11.0 进行蛋白互作(PPI)网络构建;使用Cytoscape 3.7.1 软件进行GO和KEGG富集分析;通过体内实验检测各组大鼠血糖及血脂指标:空腹血糖(FBG)、甘油三酯(TG)和总胆固醇(TC)水平;肾功能指标:尿素氮(BUN)、肌肝(SCR)和 24h-尿微量白蛋白(24h-mALB)的水平;氧化应激相关指标:超氧化物歧化酶(SOD)、丙二醛(MDA)和过氧化氢酶(CAT)的水平;炎症因子单核细胞趋化蛋白-1(CCL2)、白细胞介素-6(IL-6)和细胞间黏附分子-1(ICAM-1)的水平.结果 获得胡芦巴中 14 个活性成分和 171 个"胡芦巴-糖尿病肾病"的交集靶点,筛选出 2 个与调节炎症反应相关的核心模块,基因本体(GO)、京都基因与基因组百科全书(KEGG)富集分析表明糖基化终末产物-糖基化终末产物受体(AGE-RAGE)信号通路可能是治疗糖尿病肾病的关键通路,体内实验发现胡芦巴能对与AGE-RAGE信号通路相关的炎症因子和氧化应激反应指标起到调节作用.结论 胡芦巴可通过抗炎和抗氧化应激实现对AGE-RAGE信号通路的调控并发挥对糖尿病肾病的治疗作用.

Objective To explore the anti-inflammatory and antioxidant effects of fenugreek on diabetes nephropathy through network pharmacological analysis and in vivo experimental verification.Methods TCMSP,SwissTarget and UniProt databases were used to screen,supplement and correct the active components of fenugreek and related targets.GeneCard,DisGNet,DigSee,TTD,OMIM,and DrugBank databases were used to obtain relevant targets for diabetic nephropathy;PPI network construction with STRING 11.0;GO and KEGG enrichment analysis were performed using Cytoscape 3.7.1 software;Blood glucose and blood lipid indexes of each group were detected by in vivo experiment,including FBG,TG and TC levels.Renal function indicators:BUN,SCR and 24h-mALB levels;Oxidative stress related indexes:SOD,MDA and CAT levels;Levels of inflammatory cytokines CCL2,IL-6 and ICAM-1.Results 14 active ingredients and 171 intersecting targets of"fenugreek-diabetic nephropathy"in fenugreek were obtained,and 2 core modules related to the regulation of inflammatory response were screened,and GO and KEGG enrichment analysis showed that the AGE-RAGE signaling pathway may be the key pathway for the treatment of diabetic nephropathy,and the AGE-RAGE signaling pathway was related to inflammatory factors and oxidative stress responses.Conclusion Fenugreek can regulate the AGE-RAGE signaling pathway through anti-inflammatory and antioxidative stress and play a therapeutic role in diabetic nephropathy.
