Tiam1差异表达对卵巢癌预后及其生物学功能的影响OA
Impact of differential expression of Tiam1 on prognosis and biological functions of ovarian cancer
目的 评估Tiam1在卵巢癌中的差异表达及临床价值,探讨Tiam1对卵巢癌细胞生物学功能的影响.方法 利用cBioportal、Sangerbox等数据库分析Tiam1在多种肿瘤中的mRNA改变及表达水平;运用生物信息学分析其在卵巢癌组织中的表达及其预后.采用Linkedomics数据库分析Tiam1对卵巢癌下游基因及相关通路的影响;采用免疫组化SP法检测Tiam1蛋白在卵巢组织中的表达;构建Tiam1差异表达稳转卵巢癌细胞株,明确Tiam1差异表达对卵巢癌细胞增殖、迁移及上皮-间充质转化(epithelial-mesenchymal transition,EMT)进程的影响.结果 多个数据库检索发现Tiam1在卵巢癌等多种肿瘤组织中的mRNA表达均高于正常组织(P<0.05),且Tiam1高表达与卵巢癌的恶性程度、不良预后及化疗药物敏感性密切相关(P<0.05).免疫组化染色进一步证实Tiam1在卵巢癌中阳性表达或强阳性表达,在卵巢良性病变中不表达(P<0.05).Linkedomics数据库富集分析结合细胞功能实验共同证实Tiam1表达与细胞迁移呈显著相关,且促进卵巢癌细胞的EMT进程.结论 Tiam1高表达预示卵巢癌患者的不良预后及其对化疗药物的响应,并促进卵巢癌细胞的增殖、迁移及EMT进程,有望成为卵巢癌患者靶向治疗的新靶点.
Objective To investigate the expression levels and clinical value of Tiam1 in ovarian cancer,and to unveil its effects on the biological processes of ovarian cancer cells.Methods Databases such as cBioPortal and SangerBox were used to analyze the mRNA alterations and expression levels of Tiam1 across various tumor types.Bio-informatics analyses were conducted to investigate Tiam1 expression in ovarian cancer tissues and its correlation with patient prognosis.The Linkedomics database was utilized to identify Tiam1-regulated downstream genes and associ-ated signaling pathways in ovarian cancer.Immunohistochemical SP staining was performed to detect the protein ex-pression of Tiam1 in ovarian tissues.Stable ovarian cancer cell lines with differential Tiam1 expression were con-structed to assess the effects of Tiam1 on cell proliferation,migration,and epithelial-mesenchymal transition(EMT)process.Results Multi-database analyses revealed that Tiam1 mRNA expression levels were significantly higher in various tumor tissues including ovarian cancer than in normal tissues(P<0.05).Additionally,high Tiam1 expression was closely associated with the increased malignancy,poor prognosis,and altered chemotherapeutic drug sensitivity in ovarian cancer patients(P<0.05).Immunohistochemical staining further confirmed that Tiam1 protein showed posi-tive expression in ovarian cancer tissues and negative expression in benign ovarian lesions(P<0.05).Both enrich-ment analysis based on the Linkedomics database and in vitro cell function assays consistently demonstrated that Tiam1 expression was significantly associated with enhanced cell migration and promoted the EMT process in ovarian cancer cells.Conclusion High Tiam1 expression predicts poor prognosis and increased sensitivity to chemotherapeutic drugs.Additionally,Tiam1 promotes cancer cell proliferation,migration,and EMT process in ovarian cancer cells,making it a promising novel biomarker for targeted therapy in ovarian cancer patients.
