与天然抗性相关的巨噬蛋白(NRAMP)是一类广泛存在于植物中的金属离子转运蛋白,主要参与Cd2+、Mn2+等二价金属离子的运输.为探究芝麻NRAMP家族的结构与功能,本研究基于芝麻全基因组数据,利用生物信息学方法鉴定出SiNRAMP家族成员,并系统分析其蛋白质和基因的结构特征,以及重金属胁迫下的表达模式.结果表明,SiNRAMP家族家族共包含 6 个成员,分布于 5 条染色体上,其氨基酸长度为447~553,所有 SiN-RAMP家族成员均定位于细胞膜.所有SiNRAMP基因启动子区域均含有逆境胁迫响应元件及多种激素响应元件.SiNRAMP家族成员对不同重金属离子胁迫的响应存在差异.Cd2+处理 72 h,SiNRAMP2、SiNRAMP3、SiNRAMP5、SiNRAMP6 表达量均显著高于对照(P<0.05),表明这些 NRAMP 基因共同参与重金属的跨膜转运.此外,SiN-RAMP3 可同时响应Cd2+与Cu2+胁迫,表明其可能具有多种重金属离子转运功能.本研究为深入解析芝麻NRAMP基因的生物学功能及其在重金属胁迫响应中的分子机制提供了理论依据.

Natural resistance-associated macrophage proteins(NRAMP)are a class of metal ion transporters widely present in plants,mainly involved in the transport of divalent metal ions such as Cd2+and Mn2+.To explore the structure and function of the NRAMP family in Sesamum indicum L.,this study identified SiNRAMP family members using bioinfor-matics methods based on sesame whole-genome data,and systematically analyzed their protein and gene structural charac-teristics,as well as expression patterns under heavy metal stress.The results showed that the SiNRAMP family contained a total of six members,distributed on five chromosomes,with amino acid lengths ranging from 447 to 553.All SiNRAMP family members were localized to the cell membrane.The promoter regions of all SiNRAMP genes contained stress-responsive elements and various hormone-responsive ele-ments.The responses of SiNRAMP family members to dif-ferent heavy metal ion stresses were diverse.After 72 h of Cd2+treatment,the expression levels of SiNRAMP2,SiN-RAMP3,SiNRAMP5,and SiNRAMP6 were significantly higher than those in the control group(P<0.05),indicating that these NRAMP genes were collectively involved in the transmembrane transport of heavy metals.In addition,SiNRAMP3 could respond to both Cd2+and Cu2+stresses,suggesting its potential ability to transport multiple heavy metal ions.This study provides a theoretical basis for further elucidating the biological functions of sesame NRAMP genes and their molecular mechanisms in response to heavy metal stress.
