本研究通过RT-PCR技术从甘薯[Ipomoea batatas(L.)Lam.]中克隆获得IbEFR基因,该基因编码序列(CDS)全长3 108 bp,可编码1 035个氨基酸残基.结构域分析结果显示,IbEFR蛋白含有丰富的亮氨酸重复序列(LRR)及丝氨酸/苏氨酸激酶(S_TKc)保守结构域;序列比对结果表明,IbEFR蛋白与拟南芥AtEFR蛋白的氨基酸序列一致性为44.76%;系统进化树分析结果表明,IbEFR基因与烟草EFR基因的亲缘关系最近.理化性质预测结果表明,IbEFR蛋白理论相对分子量约为112 605,理论等电点为 5.8;氨基酸组成中,亮氨酸含量为 14.8%,丝氨酸含量为10.0%,甘氨酸含量为8.1%,分别位列前3位;不稳定指数为36.60,由此判定其为稳定型蛋白质.磷酸化位点预测结果显示,IbEFR蛋白的丝氨酸位点数最多(64 个),其次为苏氨酸位点数(19 个).亲疏水性预测结果表明,IbEFR为亲水性蛋白质;跨膜结构域预测结果显示,IbEFR蛋白含2个明显的跨膜结构域;亚细胞定位预测结果显示,IbEFR蛋白定位在细胞质的可能性最大;二级结构预测结果表明,IbEFR蛋白以无规则卷曲为主,其次为α-螺旋和β-折叠;表达模式分析结果显示,IbEFR基因可被脱落酸(ABA)、茉莉酸(JA)和水杨酸(SA)诱导上调表达,表明其可能参与甘薯的激素信号调控途径.本研究结果为后续深入解析IbEFR基因功能奠定了基础.

In this study,the IbEFR gene was cloned from Ipomoea batatas(L.)Lam.using RT-PCR technology.This gene has a full-length coding sequence(CDS)of 3 108 bp,encoding a protein of 1 035 amino acid residues.Domain analysis results showed that the IbEFR protein contained abundant leucine-rich repeat(LRR)sequences and a conserved serine/threonine kinase(S_TKc)domain.Sequence alignment results indicated that the amino acid sequence identity between the IbEFR protein and the Arabidopsis thaliana AtEFR protein was 44.76%.Phylogenetic tree analysis results revealed that the IbEFR gene had the closest genetic relationship with the EFR gene of tobacco.The prediction results of physicochemical properties indicated that the theoretical relative molecular mass of the IbEFR protein was approximately 112 605,and its theoretical isoelectric point was 5.8.In terms of amino acid composition,leucine content was 14.8%,serine con-tent was 10.0%,and glycine content was 8.1%,ranking as the top three,respectively.The instability index was 36.60,so it was determined to be a stable protein.Phosphorylation site prediction results showed that the IbEFR protein had the most serine sites(64),followed by threonine sites(19).Hydrophilicity and hydrophobicity prediction results indicated that IbEFR was a hydrophilic protein.Transmembrane domain prediction results showed that the IbEFR protein contained two obvious transmembrane domains,and the subcellular localiza-tion was most likely in the cytoplasm.Secondary structure prediction results demonstrated that the IbEFR protein was mainly composed of random coils,followed by α-helices and β-sheets.Expression pattern analysis results showed that the expression of the IbEFR gene could be induced and upregulated by abscisic acid(ABA),jasmonic acid(JA)and salicylic acid(SA),indicating that it may be involved in the hormone signal regulation pathway of sweet potato.The results of this study lay a foundation for the subsequent in-depth analysis of the function of the IbEFR gene.
