靶向DPP4多靶点干预策略治疗糖尿病的研究进展OA

Research advances in multi-target intervention strategies for DPP4 inhibition in diabetes management

二肽基肽酶-4(DPP4)抑制剂在2型糖尿病治疗中具有良好的安全性和有效性,但随着患者病程延长,其降糖效果逐渐减弱.基于DPP4 抑制剂的药物联用策略可以显著地提升血糖控制水平,而针对多靶点(如DPP4/SGLT2)的单分子调控剂则为糖尿病治疗提供了新方向.目前虽已有若干多靶点候选化合物报道,但尚无药物上市,其协同机制与成药性仍需深入评估.本研究根据联用靶点类型对DPP4抑制剂联合用药策略进行综述,并探讨相关多靶点小分子调控剂的研究进展.

Despite favorable tolerability and efficacy in type 2 diabetes,the glycemic control achieved with DPP4 inhibitors is often compromised by disease progression.The drug combination strategy based on DPP4 inhibitors has demonstrated significant potential in improving glycemic control,while multi-target small molecule regulators(such as dual DPP4/SGLT2 inhibitors)offer a promising new direction for diabetes therapy.Although several multi-target candidate compounds have been reported to date,none has been progressed to market approval.The underlying synergistic mechanisms and overall drug-likeness of these compounds require further investigation.This review summarizes the current combination therapy strategies involving DPP4 inhibitors according to the types of co-targets and discusses recent advances in the development of multi-target small molecule regulators.
