碘帕醇注射液对小鼠睾酮合成与分泌的影响OA
Effects of iopamidol injection on testosterone synthesis and secretion in mouse
为研究碘帕醇注射液对小鼠睾丸组织睾酮合成与分泌的影响,将25只雄性ICR小鼠随机分为对照组(C-24)和碘帕醇24h(I-24)、48h(I-48)、72h(I-72)、96h(I-96)组,碘帕醇组尾静脉注射500 μL碘帕醇注射液,对照组注射等量的生理盐水,监测体重及行为变化;采用ELISA试验检测小鼠血清中睾酮的浓度;采用实时荧光定量PCR和Western blot试验检测小鼠睾丸中睾酮合成关键酶(StAR、CYP11A1、3β-HSD)基因和蛋白质的表达情况;通过HE染色观察各组小鼠睾丸组织的形态变化.结果显示,各处理组小鼠体重变化均不明显,行为反应无明显异常;与对照组相比,小鼠血清睾酮浓度于碘帕醇注射后24和48 h显著降低(P<0.05),而注射后72和96 h,血清睾酮浓度有所降低,差异不显著;StAR、CYP11A1、3β-HSD基因及蛋白质表达水平与对照组相比,在碘帕醇注射24和48 h后均显著下降(P<0.05),且随着时间推移(72、96 h)其表达量逐渐恢复至接近对照值;各处理组小鼠睾丸组织结构相对完整,生精小管发育良好,各级生精细胞排列紧密有序.综上,本研究所选剂量的碘帕醇注射液注射后能暂时降低雄性小鼠睾酮的合成与分泌,但这种影响随着时间的推移会逐渐减轻,为碘帕醇注射液的临床应用及风险评估提供了试验依据.
To investigate the effects of iopamidol injection on testosterone synthesis and secretion in mice testicular tis-sues,twenty-five male ICR mice were randomly assigned to the following groups:a control group(C-24)and iopamidol groups at 24 h(I-24),48 h(I-48),72 h(I-72),and 96 h(I-96).Mice in the iopamidol groups received a tail vein injec-tion of 500 μ L iopamidol injection,while the control group received an equal volume of physiological saline.Body weight and behavioral changes were monitored throughout the experimental period.Testosterone concentration in mice serum was detected by ELISA;the expression of genes and proteins of key enzymes of testosterone synthesis(StAR,CYP11A1,3β-HSD)in mice testis was detected by real-time fluorescence quantitative PCR and Western blot;and the changes of testicu-lar tissue morphology in each group of mice were observed by HE staining.The results demonstrated that body weight changes in all treatment groups were not significant,and no obvious abnormalities in behavioral responses were observed.Serum testosterone concentrations decreased significantly at 24 and 48 h after iopamidol injection(P<0.05)compared with the control group.However,at 72 and 96 h post-injection,serum testosterone concentrations showed a decreasing trend,but the differences were not statistically significant.Similarly,the mRNA and protein expression levels of StAR,CYP11A1,and 3β-HSD were significantly lower than those in the control group at 24 and 48 h after iopamidol injection(P<0.05).Nevertheless,their expression levels gradually recovered to values close to the control group over time(72 and 96 h).Furthermore,the testicular tissue structure in all treatment groups remained relatively intact,with well-devel-oped seminiferous tubules and tightly ordered arrangement of spermatogenic cells at various stages.Based on the findings above,it can be concluded that the selected dose of iopamidol injection in this study can temporarily reduce testosterone synthesis and secretion in male mice after injection.However,this effect subsides gradually over time.These results pro-vide experimental evidence for the clinical application and risk assessment of iopamidol injection.
