外源尿囊素对碱胁迫下菜豆种子萌发及抗性的影响OA

为探究尿囊素对碱胁迫环境下菜豆种子萌发及生理特征的影响,本研究以碱敏感菜豆品种金冠为试验材料,在筛选敏感碱胁迫浓度和适宜尿囊素浸种浓度的基础上,设置蒸馏水浸种+蒸馏水萌发培养对照(CK)、蒸馏水浸种+40 mmol/L NaHCO3萌发培养处理(S)、2 mmol/L尿囊素浸种+40 mmol/L NaHCO3萌发培养处理(S+All),测定培养后 12 h、24 h、36 h、48 h、60 h和 72 h菜豆种子中可溶性蛋白质、可溶性糖、脯氨酸等渗透调节物质的含量,菜豆子叶中超氧化物歧化酶(SOD)、过氧化物酶(POD)及过氧化氢酶(CAT)等抗氧化酶的活性及超氧阴离子自由基(O2·-)产生速率、丙二醛(MDA)含量.结果表明,2 mmol/L尿囊素浸种缓解了菜豆种子萌发的碱胁迫,发芽势、发芽率分别较蒸馏水处理提高了 17.71%、17.76%.培养后12~72hS处理菜豆种子的可溶性蛋白质含量、可溶性糖含量、脯氨酸含量及菜豆子叶中的O2·-产生速率、MDA含量总体高于CK,而菜豆子叶的抗氧化酶活性总体低于CK.S+All处理菜豆种子的可溶性蛋白质含量、可溶性糖含量、脯氨酸含量及菜豆子叶的抗氧化酶活性总体高于S处理,而菜豆子叶的O2·-产生速率和MDA含量总体低于S处理.本研究结果为尿囊素在碱胁迫环境下菜豆种植中的应用提供了依据.

To explore the effects of allantoin on seed germination and physiological characteristics of common bean under alkali stress,the alkali-sensitive common bean variety Jinguan was used as the experimental material in this study.Based on the screening of sensitive alkali stress concentration and suitable allantoin soaking concentration,distilled water soaking+distilled water germination culture control(CK),distilled water soaking+40 mmol/L NaHCO3 germination cul-ture treatment(S),2 mmol/L allantoin soaking+40 mmol/L NaHCO3 germination culture treatment(S+All)were set.The contents of soluble protein,soluble sugar,proline and other osmotic adjustment substances in com-mon bean seeds,the activities of antioxidant enzymes such as superoxide dismutase(SOD),peroxidase(POD)and catalase(CAT),the production rate of superoxide anion free radical(O2·-)and the content of malondialdehyde(MDA)in cotyledons of common bean seeds were meas-ured at 12 h,24 h,36 h,48 h,60 h and 72 h after cul-ture.The results showed that the germination potential and germination rate of seeds treated with 2 mmol/L allantoin were 17.71%and 17.76%higher than those of distilled water treatment,respectively,which alleviated alkaline stress during the germination of common bean seeds.The soluble protein content,soluble sugar content,proline content in common bean seeds,and O2·-production rate and MDA content of the cotyledons of bean seeds treated with S at 12-72 h after culture were generally higher than those of CK,while the antioxidant enzyme activity was generally lower than that of CK in cotyle-dons of common bean.The soluble protein content,soluble sugar content,proline content in common bean seeds and an-tioxidant enzyme activity in cotyledons of common bean of S+All treatment were higher than those of S treatment,while the O2·-production rate and MDA content in cotyledons of common bean were lower than those of S treatment.The results of this study provide a basis for the application of allantoin in bean planting under alkaline stress.
