为研究阿如拉-7味散(Arula-7 powder,AR)对致病性大肠杆菌(Enteropathogenic Escherichia coli,EPEC)感染引起的大鼠肠道损伤的修复作用,本试验将 50 只SD大鼠随机分为 5 组(分别命名为BC组、NC组、PC组、ARH组、ARL组),每组 10 只.BC组为空白对照,不做任何处理;NC组为阴性对照,灌服灭菌生理盐水+腹腔注射致病性大肠杆菌;PC组为阳性对照,灌服环丙沙星(用量为 0.13 g/kg)+腹腔注射致病性大肠杆菌;ARH组灌服阿如拉-7味散(用量为 1.08 g/kg)+腹腔注射致病性大肠杆菌;ARL组灌服阿如拉-7 味散(用量为 0.54 g/kg)+腹腔注射致病性大肠杆菌.结果显示,NC组大鼠盲肠乳酸菌和双歧杆菌数量显著低于BC组,NC组肠杆菌和肠球菌数量显著高于BC组;NC组肠组织中的TNF-α含量、IL-6 含量和IL-1β含量显著高于BC组,IFN-γ含量则显著低于BC组;NC组大鼠回肠黏膜紧密连接(TJ)超微结构明显损伤、桥粒(DE)结构异常,微绒毛杂乱且有断裂现象;与BC组相比,NC组紧密连接蛋白基因Occludin、Claudin-1和ZO-1相对表达量显著下降,且细菌移位情况严重;ARH组与NC组相比,肠杆菌和肠球菌数量显著降低,乳酸菌和双歧杆菌数量显著提高,肠组织中IL-1β、IL-6 和TNF-α含量显著降低,TJ结构明显趋于正常,紧密连接蛋白基因Occludin、Claudin-1 和ZO-1 相对表达量显著提高(P<0.05),且细菌移位现象明显减少.由此可见,高剂量的阿如拉-7 味散可通过调节大鼠肠道菌群结构、改善肠上皮细胞紧密连接结构以及抑制炎症反应对EPEC感染导致的肠道损伤起到修复作用.

To investigate the repairing effect of Arula-7 powder(AR)on intestinal injury in rats induced by entero-pathogenic Escherichia coli(EPEC)infection,fifty SD rats were randomly divided into blank control(BC)group,negative control(NC)group,positive control(PC)group,ARH group and ARL group,with ten rats in each group.The BC group received no treatment,whereas the NC,PC,ARH,and ARL groups were intragastrically administered sterile normal saline,0.13 g/kg ciprofloxacin,1.08 g/kg Arula-7 powder,and 0.54 g/kg Arula-7 powder,respectively,and were intraperitoneally injected with EPEC.The results showed that the counts of lactobacilli and bifidobacteria in the cecum of the NC group were significantly lower than those in the BC group,while the counts of enterobacteria and enterococci in the NC group were significantly higher than those in the BC group.The levels of TNF-α,IL-6,and IL-1β in the intestinal tissue of the NC group were significantly higher than those in the BC group,while the IFN-γ level was significantly lower.In the NC group,the ultrastructure of ileal mucosal tight junctions(TJ)was markedly damaged,desmosome(DE)structures were abnormal,and the microvilli ap-peared disordered and fragmented.Compared with the BC group,the relative expression levels of tight junction protein genes Occludin,Claudin-1,and ZO-1 in the NC group were significantly decreased,and severe bacterial translocation was observed.Compared with the NC group,the ARH group showed a significant decrease in the counts of enterobacteria and enterococci,and a significant increase in the counts of lactobacilli and bifidobacteria.The levels of IL-1β,IL-6,and TNF-α in intestinal tissue were significantly reduced,the tight junction structure was markedly improved toward normal,and the relative expression levels of the tight junction protein genes Occludin,Claudin-1,and ZO-1 were significantly upregulated(P<0.05).Additionally,bacterial translocation was noticeably reduced.Therefore,high-dose Arula-7 powder exerts a re-pairing effect on intestinal damage induced by EPEC infection by modulating the gut microbiota structure,improving the tight junction structure of intestinal epithelial cells,and suppressing inflammatory responses.
