目的 研究丹夏调斑方对氧化低密度脂蛋白(low density lipoprotein,ox-LDL)诱导的人脐静脉内皮细胞(human umbilical vein endothelial cells,HUVECs)铁死亡的影响,一定程度上揭示丹夏调斑方防治动脉粥样硬化的机制.方法 采用150 μg/mL的ox-LDL诱导HUVECs建立体外动脉粥样硬化(atherosclerosis,AS)模型,将其分为空白对照组、ox-LDL模型组、ox-LDL+阴性血清对照组、ox-LDL+含药血清低剂量组、ox-LDL+含药血清中剂量组、ox-LDL+含药血清高剂量组、铁死亡抑制剂组.采用透射电镜观察细胞线粒体形态;试剂盒检测细胞Fe2+含量;流式细胞术检测细胞凋亡率及细胞内和脂质活性氧(reactive oxygen species,ROS)水平.结果 与ox-LDL模型组相比,丹夏调斑方含药血清组和铁死亡抑制剂组细胞中Fe2+、细胞内ROS和脂质ROS水平显著降低(P<0.05,P<0.01)细胞存活率水平显著升高(P<0.05,P<0.01).与空白对照组相比,ox-LDL模型组细胞在透射电镜下可见线粒体形态变小,且明显皱缩;丹夏调斑方含药血清低、中、高血清组能显著阻止线粒体皱缩,且呈剂量依赖性.结论 ox-LDL诱导的HUVECs中发生了铁死亡,丹夏调斑方含药血清能够抑制铁死亡,发挥对ox-LDL诱导的HUVECs的保护作用.

Objective To investigate the effect of Danxia Tiaoban Decoction on ferroptosis in human umbilical vein endothelial cells(HUVECs)induced by oxidized low-density lipoprotein(ox-LDL),and to reveal to some extent the mechanism of Danxia Tiaoban Decoction in preventing and treating athero-sclerosis(AS).Methods HUVECs were induced by 150 μg/mL ox-LDL to establish an in vitro AS model,which was divided into the blank group,the model group,the negative serum group,the low,medium and high concentration of Danxia Tiaoban Decoction serum groups,and the ferroptosis inhibitor group.Transmission electron microscopy was used to observe the morphology of mitochondria;kits were used to detect the Fe2+content in cells,and flow cytometry was used to detect the apoptosis rate of cells and the levels of intracellular and lipid ROS.Results After induction by ox-LDL,compared with the model group,the levels of Fe2+,intracellular ROS and lipid ROS,and cell survival rate in the Danxia Tiaoban Decoction serum groups and the ferroptosis inhibitor group were significantly increased(P<0.05,P<0.01);compared with the blank group,the mitochondria in the model group were smaller and significantly shrunken under transmission electron microscopy,while the low,medium and high concentration Danxia Tiaoban Decoction serum groups could significantly prevent mitochondrial shrinkage in a dose-dependent manner.Conclusion Ferroptosis occurred in HUVECs induced by ox-LDL,and Danxia Tiaoban Decoction serum could inhibit ferroptosis and exert a protective effect on HUVECs induced by ox-LDL.
