化浊解毒方调控线粒体动力学保护慢性肾衰竭大鼠肾功能的作用研究OA

Study on the effect of Huazhuo Jiedu Decoction on regulating mitochondrial dynamics and protecting renal function in CRF rats

目的 研究化浊解毒方通过调控肾脏线粒体动力学保护慢性肾衰竭(chronic renal failure,CRF)大鼠肾功能的作用机制.方法 采用5/6 肾切除法制备CRF大鼠模型,67 只SD大鼠随机选取11 只为假手术组和56 只造模组,将造模成功的SD大鼠随机分为模型组、化浊解毒方低剂量组(1.175 g/kg)、化浊解毒方中剂量组(2.35 g/kg)、化浊解毒方高剂量组(4.7 g/kg)、氯沙坦钾组(4.5×10-3 g/kg),每组9 只.各给药组分别予相应药物灌胃,假手术组和模型组予等量生理盐水,连续给药8 周.苏木素—伊红(hematoxylin-eosin staining,HE)染色和马松(Masson's trichrome stain,Masson)染色观察肾脏组织病理变化;透射电镜观察肾小管上皮细胞线粒体超微结构变化;蛋白免疫印迹法(western blot,WB)检测动力相关蛋白1(dynamin-related protein 1,Drp1)、线粒体融合蛋白1(mitofusin 1,Mfn1)、线粒体融合蛋白 2(mitofusin 2,Mfn2)、Bcl-2 相关 X 蛋白(Bcl-2-associated X protein,Bax)、B细胞淋巴瘤-2(B-cell lymphoma 2,Bcl-2)蛋白表达水平;免疫组化法检测Drp1、Mfn1、Mfn2 蛋白表达水平;比色法检测肾脏三磷酸腺苷(adenosine triphosphate,ATP)、二磷酸腺苷(adenosine diphosphate,ADP)、单磷酸腺苷(adenosine monophosphate,AMP)值水平.结果 与假手术组比较,模型组大鼠部分肾小球萎缩,系膜细胞及基质增生,肾小管间质区域结构紊乱,小管刷毛缘脱落,空泡样变性,部分肾小管萎缩,球周及间质大量蓝色胶原纤维堆积;电镜下可见肾小管上皮细胞线粒体肿胀、嵴断裂、空泡样变;Drp1、Bax蛋白表达水平明显升高(P<0.01),Mfn1、Mfn2、Bcl-2蛋白表达水平明显降低(P<0.01);ATP、ADP、AMP含量明显降低(P<0.01).经化浊解毒方干预后,大鼠肾脏上述病理损伤明显改善;电镜下可见肾小管上皮细胞线粒体结构损伤亦明显减轻;Drp1、Bax蛋白表达水平明显下降(P<0.05,P<0.01),Mfn1、Mfn2、Bcl-2 蛋白表达水平明显升高(P<0.05,P<0.01);ATP、ADP、AMP值明显升高(P<0.05,P<0.01).结论 化浊解毒方能有效改善5/6 肾切除大鼠肾功能,可能与其降低Drp1 蛋白水平,升高Mfn1、Mfn2 蛋白表达水平,改善线粒体动力学,增加ATP产能,抑制细胞凋亡,减轻肾脏纤维化相关.

Objective To study the mechanism of Huazhuo Jiedu Decoction protecting renal function in rats with chronic renal failure(CRF)by regulating renal mitochondrial dynamics.Methods The rat model of chronic renal failure(CRF)was established by 5/6 nephrectomy.11 of 67 SD rats were randomly selected as the sham operation group and 56 as the model group,and the successfully modeled SD rats were divided into the model group,the Huazhuo Jiedu Decoction low-dose group(1.175 g/kg),the Huazhuo Jiedu Decoction medium-dose group(2.35 g/kg),the Huazhuo Jiedu Decoction high-dose group(4.7 g/kg),the losartan potassium group(4.5×10-3 g/kg),9 rats in each group.The administration groups were given the corresponding drugs by gavage,and the sham operation group and the model group were given the same amount of normal saline for 8 weeks.The pathological changes of renal tissue were observed by hematoxylin-eosin(HE)staining and Masson's trichrome stain(Masson)staining.The ultra-structural changes of mitochondria in renal tubular epithelial cells were observed by transmission electron microscopy.The expression levels of dynamin-related protein 1(Drp1),mitochondrial fusion protein 1(Mfn1),mitochondrial fusion protein 2(Mfn2),Bcl-2-related X protein(Bax)and B-cell lymphoma-2(Bcl-2)were detected by western blot(WB).The expression levels of Drp1 and Mfn2 proteins were detected by immunohistochemistry.The levels of adenosine triphosphate(ATP),adenosine diphosphate(ADP)and adenosine monophosphate(AMP)in kidney were detected by colorimetry.Results Compared with the sham operation group,some glomeruli atrophy,mesangial cells and matrix hyperplasia,structural disorder of renal tubulointerstitial area,tubular brush border shedding,vacuolar degeneration,partial renal tubular atrophy,and a large number of blue collagen fibers accumulated in periglomerular and interstitial tissues were showed in the model group.Renal tubular epithelial cells mitochondrial swelling,ridge fracture,vacuolar degeneration.The expression levels of Drp1 and Bax protein were significantly increased(P<0.01),and the expression levels of Mfn1、Mfn2 and Bcl-2 protein were significantly decreased(P<0.05,P<0.01).The contents of ATP,ADP and AMP were significantly decreased(P<0.01).After the intervention of Huazhuo Jiedu Decoction,the above pathological damage was significantly improved,mi-tochondrial structure damage of renal tubular epithelial cells was reduced.The expression levels of Drp1 and Bax protein were significantly decreased(P<0.05,P<0.01),and the expression levels of Mfn1、Mfn2 and Bcl-2 protein were significantly increased(P<0.05,P<0.01).The values of ATP,ADP and AMP were significantly increased(P<0.05,P<0.01).Conclusion Huazhuo Jiedu Decoction can effectively improve the renal function of 5/6 nephrectomy rats.This renal protective effect may be related to the reduction of Drp1 protein level and the increase of Mfn1、Mfn2 protein expression level,thereby improving mitochondrial dynamics,increasing ATP production,inhibiting apoptosis and reducing renal fibrosis.
