自然群体小麦旗叶和株高的全基因组关联分析OA
Genome-wide Association Study of Flag Leaf and Plant Height in a Natural Wheat Population
[目的]探明小麦自然群体中与旗叶和株高性状显著相关的遗传位点,为挖掘小麦优异基因及遗传育种提供理论依据.[方法]以 300份小麦自然群体为材料,结合 120K液相芯片进行基因分型,在 2年3个环境下测定旗叶长、旗叶宽和株高,利用混合线性模型进行全基因组关联分析,筛选与小麦旗叶、株高性状相关的候选基因.[结果]共定位到 28个与小麦旗叶长、旗叶宽和株高显著关联的SNP位点,有 4个位点与旗叶性状相关(分布于 2B、4B、5B、6A染色体),24个与株高相关(分布于 1A、1B、1D等 11条染色体).旗叶长在 5B染色体位点(490 218 405~490 219 967 bp)注释到与氧依赖性FAD关联氧化还原酶家族相关的基因,旗叶宽在2B染色体位点(721 411 051~721 413 048 bp)、4B染色体位点(608 452 172~608 452 558 bp)分别注释到与细胞色素P450、锌指结构相关的基因.株高相关位点中,在4B染色体区间(652 574 813~652 575 145 bp)注释到核糖核酸酶H蛋白基因TraesCS4B03G0941800LC,可能参与株高矮化调控.[结论]共检测出 28个小麦旗叶和株高的关键遗传位点,鉴定出潜在新基因TraesCS4B03G0941800LC,可为小麦矮化育种提供新的分子标记.
[Objective]The genetic loci significantly related to flag leaf and plant height in a natural wheat population were investigated to provide the theoretical basis for development of excelllent genes and genetic breeding in wheat.[Method]The genotyping of 300 wheat varieties(lines)was analyzed by using a 120K SNP liquid chip.The flag leaf length(FLL),flag leaf width(FLW)and plant height(PH)of wheat plants in a natural population composed of 300 wheat varieties(lines)were determined under three different environments in 2023 and 2024.The genome-wide association study(GWAS)of flag leaf and plant height was conducted by a mixed linear model(MLM)to screen candidate genes.[Result]A total of 28 SNP loci significantly associated with wheat FLL,FLW,and PH were identified.Among them,four loci associated with flag leaf traits were located on chromsome 2B,4B,5B and 6A,and 24 SNP loci associated with plant height were located on 11 chromosomes such as 1A,1B,1D and so on.For flag leaf length,a gene related to the oxygen-dependent FAD-linked oxidoreductase family was annotated near the locus on chromosome 5B(490 218 405-490 219 967 bp).For flag leaf width,genes encoding cytochrome P450 and a zinc finger protein were annotated near the loci on chromosome 2B(721 411 051-721 413 048 bp)and chromosome 4B(608 452 172-608 452 558 bp),respectively.Among the loci related to plant height,the ribonuclease H protein gene TraesCS4B03G0941800LC was annotated in the 4B chromosome interval(652 574 813-652 575 145 bp),which may be involved in the dwarfing regulation of plant height.[Conclusion]28 key genetic loci related to flag leaf traits and plant height are located,and a new potential gene(TraesCS4B03G0941800LC)related to plant height is identified to provide a new molecular marker for dwarfing breeding in wheat.
