[目的]探明贵州古茶树种质的表型多样性,为贵州茶树种质资源挖掘与保护提供理论依据.[方法]以贵州望谟、普安和务川 66份古茶树种质为研究对象,采用超景深三维显微镜拍照、直接观察表型等方法调查形态特征与叶片表型性状,并进行遗传多样性分析、主成分分析和聚类分析.[结果]普安和务川古茶树种质为乔木型,望谟种质有小乔木和灌木 2种类型;普安种质顶芽和子房均无毛,务川种质顶芽和子房存在有毛和无毛 2种性状,望谟种质顶芽和子房均有毛.66份古茶树种质叶片质量性状的遗传多样性指数为 0.517~1.032,均值为 0.788,表现为叶片大小>叶缘>叶片质地>叶齿锐度>叶身>叶齿深度>叶尖>叶形>叶齿密度>叶面>叶色>叶基;数量性状的变异系数为 11.25%~17.43%,均值为 15.21%,遗传多样性指数为 1.399~4.022,均值为 2.837,叶长的遗传多样性指数是叶脉对数的 2.87倍.不同地区同一性状的遗传多样性指数和变异系数存在差异,普安、望谟和务川古茶树种质资源的叶片形态特征遗传多样性均较丰富,且存在明显的地域特征.主成分分析结果显示,15个叶片表型性状排名前 6位的主成分特征值>1,累计贡献率为 73.922%,可反映叶片表型性状的基本信息,即叶长、叶宽、叶片大小、叶齿深度、叶尖、叶身、叶面等性状是其形态分化的主要指标.聚类分析表明,66份古茶树种质分为 4个类群,分别有 32份、8份、8份和18份种质.类群Ⅰ多源于务川和普安,类群Ⅱ多源于普安,类群Ⅲ均源于望谟,类群Ⅳ含普安 3份、务川4份、望谟 11份种质.[结论]贵州古茶树种质资源遗传多样性丰富,各区域古茶树植物学分类存在明显差异.望谟红紫芽和红花的古茶树种质可作为红紫芽品种选育、制作红花茶的潜在利用资源;务川和普安顶芽无毛的种质可作为制作翠芽茶的原材料;务川顶芽有毛的种质可作为毛尖茶的原材料.

[Objective]The phenotypic diversity of 66 ancient tea tree germplasm resources from Guizhou was exp-lored to provide a theoretical basis for excavation and protection of tea tree germplasm resources in Guizhou.[Method]The leaf morphological characteristics and phenotypic traits of 66 ancient tea tree germplasm resources from Wangmo,Pu'an and Wuchuan County in Guizhou were investigated by ultra-depth 3D microscopy imaging,direct phenotypic observation,genetic diversity analysis,principal component analysis and clustering analysis.[Result]The germplasm resources from Pu'an and Wuchuan County were an arbor type,while the germplasm resources from Wangmo County were a small arbor type and shrub type.The apical bud and ovary of the germplasm resources from Pu'an County both were hairless.The apical bud and ovary of the germplasm resources from Wuchuan County were of hairy and hairless.The apical bud and ovary of the germplasm resources from Wangmo County both had hair.The genetic diversity index of leaf qualitative traits of 66 ancient tea tree germplasm resources ranged from 0.517 to 1.032,with an average of 0.788,and the genetic diversity index of 12 leaf qualitative traits was leaf size>leaf margin>leaf texture>sharpness of leaf serration>leaf lamina>depth of leaf serration>leaf opex>leaf shape>density of leaf serration>leaf surface>leaf color>leaf base.The variation coefficient and genetic diversity index of leaf quantitative traits were 11.25%—17.43%with an average of 15.21%and 1.399-4.022 with an average of 2.837 respectively.The genetic diversity index of leaf length was 2.87 times that of leaf vein pairs.There were differences in genetic diversity index and variation coefficient of the same trait in different regions.The leaf morphological traits of 66 ancient tea tree germplasm resources from Pu'an,Wangmo,and Wuchuan Counties showed abundant genetic diversity and obvious regional characteristics.The PCA results showed that the accumulative contribution rate of top six principal components with eigenvalues>1 reached 73.922%,which reflects the basic information of leaf phenotypic traits.Seven leaf traits of leaf length,leaf width,leaf size,depth of leaf serration,leaf opex,leaf lamina and leaf surface were the main indicators of morphological differentiation.Cluster analysis showed that 66 ancient tea tree germplasm resources were classified into four groups.There were 32,8,8 and 18 germplasm resources in Group I,Group II,Group III and Group IV respectively.The germplasm resources in Group I and Group II were mostly from Wuchuan and Pu'an,and Pu'an separately.The germplasm resources in Group III all were from Wangmo.The germplasm resources in Group IV were from Pu'an(3),Wuchuan(4)and Wangmo(11).[Conclusion]The genetic diversity of ancient tea tree germplasm resources is rich in Guizhou but there is a significant difference in botanic classification of ancient tea tree germplasm resources among different regions.The red-purple bud and red-flower germplasm resources from Wangmo can be used as the potential resources for breeding red-purple bud varieties and producing red-flower tea.The hairless apical bud germplasm resources from Wuchuan and Pu'an can be used as the raw materials for making Cuiya tea.The hairy apical bud germplasm resources from Wuchuan can be used as the raw materials for Maojian tea.
