液液萃取-固相萃取/气相色谱-串联质谱法测定人血清中21种氟代液晶单体OA
Determination of 21 Kinds of Fluorinated Liquid Crystal Monomers in Human Serum by Combining Liquid-Liquid Extraction and Solid-phase Extraction with Gas Chromatography-Tandem Mass Spectrometry
建立了一种基于液液萃取-固相萃取(LLE-SPE)结合气相色谱-串联质谱(GC-MS/MS)同时测定人血清中21种氟代液晶单体(FLCMs)的分析方法.通过保留时间和结构相似性筛选定量内标,并对仪器条件、萃取溶剂、固相萃取柱及洗脱体系进行系统优化,最终确定最佳前处理流程为:250 μL血清经浓硫酸与甲醇处理后,依次用正己烷和甲基叔丁基醚进行LLE,合并萃取液,经氮吹浓缩后,采用硅胶SPE柱净化,收集二氯甲烷洗脱液,氮吹至近干,加入20 μL甲苯复溶后进行GC-MS/MS检测,采用选择反应监测(SRM)模式检测,内标法定量.方法学验证结果表明,21种FLCMs在0.5~100 μg/L浓度范围内线性良好(r≥0.999),方法检出限和定量限分别为0.045~0.272 μg/L和0.150~0.908 μg/L,基质效应为86.7%~127.8%,平均加标回收率为92.7%~134.2%,日内与日间精密度分别为2.45%~8.62%与2.01%~13.15%.采用本方法检测122份人血清样本,共检出20种FLCMs,其中5种目标物的检出率超过50%,浓度中位数范围为0.126~5.812 μg/L.本方法的基质效应可控、准确度高、重现性好,灵敏度满足痕量分析要求,适用于大规模人群调查中多种FLCMs的同时检测.
An analytical method based on liquid-liquid extraction combined with solid-phase extraction(LLE-SPE)and gas chromatography-tandem mass spectrometry(GC-MS/MS)was developed for simultaneous determination of 21 kinds of fluorinated liquid crystal monomers(FLCMs)in human serum.Quantitative internal standards were selected based on retention time and structural similarity.Instrumental conditions,extraction solvents,SPE cartridges,and elution system were systematically optimized.The optimized sample preparation procedure involved processing 250 μL of serum with concentrated sulfuric acid and methanol,followed by two sequential LLE usingn-hexane and methyl tert-butyl ether.The combined extracts were concentrated under a nitrogen stream,purified on a silica gel SPE cartridge,and eluted with dichloromethane.The eluate was concentrated to near dryness with nitrogen,reconstituted in 20 μL of toluene,and analyzed by GC-MS/MS in selected reaction monitoring(SRM)mode with internal standard calibration.Method validation confirmed good linearity for the 21 kinds of FLCMs within the range of 0.5–100 μg/L(r≥0.999).The method achieved limits of detection(LODs)between 0.045 and 0.272 μg/L,with limits of quantification(LOQs)ranging from 0.150 to 0.908 μg/L.The matrix effect was 86.7%–127.8%,with a mean spiked recoveries of 92.7%–134.2%.The intra-day and inter-day precisions(RSD)were 2.45%–8.62%and 2.01%–13.15%,respectively.The method was applied to 122 human serum samples,resulting in the detection of 20 kinds of FLCMs.Five of these target compounds exhibited a detection frequency greater than 50%,with median concentration range of 0.126–5.812 μg/L.This method was suitable for monitoring FLCMs in human serum from the general population,demonstrating controllable matrix effects,high accuracy,stable reproducibility,and adequate sensitivity.
