以硝酸铜和谷氨酸为原料,在室温和常压条件下,在水溶液中采用一步沉淀法制备了铜-谷氨酸聚合物纳米材料(Copper-glutamic acid polymer nanoparticles,Cu-Glu CPNs)用于乙酰胆碱酯酶(Acetylcholines-terase,AChE)抑制剂类农药的检测.AChE催化乙酰胆碱(Acetylcholine,ACh)水解生成胆碱(Choline,Ch),Ch被胆碱氧化酶(Choline oxidase,ChOx)氧化产生过氧化氢(H2O2),进一步通过具有类过氧化物酶活性的Cu-Glu CPNs催化H2O2 将无色的3,3',5,5'-四甲基联苯胺(3,3',5,5'-Tetramethylbenzidine,TMB)氧化为蓝色的氧化产物,在650 nm处出现特征吸收峰.当有机磷类(Organophosphorus pesticides,OPs)或氨基甲酸酯类(Carbamate pesticides,CPs)等AChE抑制剂类农药存在时,会间接抑制TMB氧化,导致650 nm处的吸光度降低,其降低幅度与农药的浓度相关,据此实现了农药的比色检测.在最优检测条件下,OPs类农药马拉硫磷(Malathion,Mal)和CPs类农药甲萘威(Carbaryl,Car)的检出限(LOD)分别为2.14和1.29 ng/mL.将本方法应用于苹果、黄瓜、西红柿和橘子样品中Mal和Car的检测,加标回收率分别为91.6%~105.7%和87.1%~118.6%,表明其可应用于实际农产品中AChE抑制剂类农药的检测.

Copper-glutamic acid polymer nanoparticles(Cu-Glu CPNs)were prepared by one-step precipitation method from copper nitrate and glutamic acid in aqueous solution at room temperature and atmospheric pressure for detection of acetylcholinesterase(AChE)inhibitor pesticides.AChE catalyzed the hydrolysis of acetylcholine(ACh)to choline(Ch),which was oxidized by choline oxidase(ChOx)to produce hydrogen peroxide(H2O2).H2O2 was further catalyzed by Cu-Glu CPNs with peroxidase-like activity to oxidize the colorless 3,3',5,5'-tetramethylbenzidine(TMB)to a blue oxidation product,showing a distinct absorption peak at 650 nm.When organophosphorus pesticides(OPs)or carbamate pesticides(CPs)and other AChE inhibitor pesticides were present,they indirectly inhibited the oxidation of TMB,resulting in a decrease in absorbance at 650 nm.The decrease was related to the pesticide concentration,thus achieving colorimetric detection of pesticides.Under the optimal conditions,the limits of detection(LOD)for OPs pesticide malathion(Mal)and CPs pesticide carbaryl(Car)were 2.14 ng/mL and 1.29 ng/mL,respectively.This method was applied to detection of Mal and Car in apples,cucumbers,tomatoes and oranges with recovery rates ranging from 91.6%to 105.7%and 87.1%to 118.6%,respectively,indicating its potential for practical application in agricultural product testing.
