溶剂萃取/HPLC-MS/MS法快速测定1,1,2,2-四氯乙烷职业接触人群尿中二氯乙酸与三氯乙酸OA
Rapid Determination of Dichloroacetic Acid and Trichloroacetic Acid in Urine of 1,1,2,2-Tetrachloroethane Occupational Exposure Population by Liquid-Liquid Extraction/HPLC-MS/MS
建立了高效液相色谱-串联质谱(HPLC-MS/MS)测定1,1,2,2-四氯乙烷职业接触人群尿中二氯乙酸(DCA)和三氯乙酸(TCA)的快速检测方法,并用于实际样品分析.尿样中先加入盐酸溶液酸化,再加入氯化钠振荡混匀,随后加入乙酸乙酯提取,上层提取液氮吹浓缩至近干并加入15%乙腈溶液复溶,微孔滤膜过滤后进行测定.采用Waters HSS T3(2.1 mm×100 mm,1.8 µm)色谱柱分离,以乙腈-0.1%乙酸水溶液为流动相进行梯度洗脱,采用电喷雾离子源负离子(ESI-)多反应监测(MRM)模式进行检测.DCA和TCA均在2.0~100 µg/L质量浓度范围内呈线性关系,线性相关系数(r)大于0.999,检出限(LOD)为0.2 µg/L,定量下限(LOQ)为1.0 µg/L.低、中、高3个加标水平下的回收率为75.3%~104%,DCA和TCA的平均回收率分别为91.1%和86.1%,相对标准偏差(RSD)为2.1%~9.5%.采用该方法测定了普通人群、常规职业人群和职业中毒患者尿中DCA、TCA.该方法简单、快速、灵敏度高,可为1,1,2,2-四氯乙烷职业接触人群职业中毒临床检验提供技术支持.
A rapid detection method using high performance liquid chromatography-tandem mass spectrometry(HPLC-MS/MS)was established for determining dichloroacetic acid(DCA)and trichlo-roacetic acid(TCA)in urine of occupationally exposed individuals to 1,1,2,2-tetrachloroethane.The urine samples were acidified by adding hydrochloric acid solution,followed with sodium chloride being added and shaking.Subsequently DCA and TCA were extracted with ethyl acetate.The upper extract was concentrated to near dryness using nitrogen blowing,and then redissolved with 15%ace-tonitrile solution.After being filtered through a microporous membrane,they were injected into the instrument.Chromatographic separation was achieved using a Waters HSS T3(2.1 mm×100 mm,1.8 µm)column,with acetonitrile-0.1%acetic acid aqueous solution as the mobile phase for gradi-ent elution.Electrospray ionization(ESI)source and multiple reaction monitoring(MRM)mode were employed.Quantification was performed using the matrix-matched curve external standard method.Both DCA and TCA exhibited linear relationships within the mass concentration range of 2.0-100 µg/L,with linear correlation coefficients(r)higher than 0.999.The limits of detection(LODs)and the limits of quantitation(LOQs)were calculated to be 0.2 µg/L and 1.0 µg/L,respectively.The re-coveries at low,medium and high spiked concentration levels ranged from 75.3%to 104%.The aver-age recoveries of DCA and TCA were 91.1%and 86.1%,respectively,with relative standard devia-tions(RSDs)ranging from 2.1%to 9.5%.The established analytical procedure was employed for urine DCA and TCA level investigating of the general population,routine occupational population,and occupational poisoning patients.The method is simple,rapid and highly sensitive,providing a technical reference for biological monitoring and clinical testing of occupational poisoning in occupa-tionally exposed individuals to 1,1,2,2-tetrachloroethane.
