目的:观察舒胃汤调控腺苷 5'-单磷酸激活蛋白激酶(AMPK)/Unc-51 样自噬激活激酶 1(ULK1)/FUN14 结构域蛋白 1(FUNDC1)信号通路对功能性消化不良(Functional Dyspepsia,FD)大鼠线粒体自噬的影响.方法:将60 只 SPF 级雄性 SD 大鼠按照随机数字表法分为正常对照组、模型对照组、舒胃汤 3.78、7.56、15.12 g/kg组及莫沙必利1.35 mg/kg组,每组各10 只.正常对照组不予造模,造模大鼠采用改良复合病因造模(慢性束缚应激+隔日夹尾激怒+过度疲乏+饮食不调),持续21 d,造成FD肝郁脾虚证模型大鼠,造模完成后评价模型是否成功,造模成功后连续灌胃14 d,观测大鼠体质量、进食量变化;旷场试验观察大鼠移动总距离与移动次数;以黑色半固体糊灌胃检测大鼠胃排空率、小肠推进率;HE染色法检测大鼠胃窦组织病理改变;试剂盒检测胃窦组织ATP 含量;Western blot法检测胃窦组织p-AMPK/AMPK、p-ULK1/ULK1、FUNDC1、LC3II/lC3I蛋白表达;免疫荧光染色检测FD肝郁脾虚证大鼠胃窦组织中LAMP2 和TOMM20 表达及共定位水平.结果:各组胃窦组织均未见明显器质性改变;与正常对照组相比,模型对照组大鼠体质量、进食量、移动总距离和直立次数、胃排空率、小肠推进率明显降低(P<0.05 或P<0.01),胃组织ATP 含量显著降低(P<0.05),胃窦组织p-AMPK/AMPK、p-ULK1/ULK1、FUNDC1、LC3II/lC3I蛋白表达明显上调(P<0.05 或P<0.01),胃窦组织TOMM20、LAMP2 荧光强度、TOMM20 与LAMP2 共定位系数明显升高(P<0.05 或 P<0.01);与模型对照组相比,舒胃汤 3.78、7.56、15.12 g/kg组及莫沙必利组大鼠体质量、进食量、胃排空率、小肠推进率、移动总距离和直立次数明显升高(P<0.05 或P<0.01),胃组织ATP 含量明显升高(P<0.05 或P<0.01),胃窦组织p-AMPK/AMPK、p-ULK1/ULK1、FUNDC1、LC3II/lC3I蛋白表达明显下调(P<0.05 或P<0.01),胃窦组织TOMM20、LAMP2 荧光强度、TOMM20 与LAMP2 共定位系数明显降低(P<0.05 或P<0.01).结论:舒胃汤对FD肝郁脾虚证大鼠的作用机制与AMPK/ULK1/FUNDC1 通路调节线粒体自噬有关.

Objective:To observe the effect of Shuwei Decoction on mitochondrial autophagy in rats with functional dyspepsia(FD)by modulating the 5'adenosine monophosphate-activated protein kinase/ULK-51-like kinase 1/FUN14 domain-containing protein 1(AMPK/ULK1/FUNDC1)signaling pathway.Methods:Sixty SPF male SD rats were divid-ed into a blank control group,model control group,Shuwei Decoction groups at doses of 3.78 g/kg,7.56 g/kg,and 15.12 g/kg,and a Mosapride group(1.35×10-3 g/kg)using a random number table,with 10 rats in each group.Ex-cept for the blank control group,rats in the other five groups were subjected to a modified compound etiology modeling method(chronic restraint stress+alternate-day tail-clamping irritation+excessive fatigue+dietary disorders)for 21 days to establish an FD model with a syndrome of liver depression and spleen deficiency,After modeling,its success was evaluated.After successful modeling,rats were administered the respective interventions by gavage for 14 days.Changes in body weight and food intake of the rats were observed.In the open-field test,the total movement distance and frequen-cy were observed.A black semi-solid paste was administered by gavage to detect the gastric emptying rate and small in-testinal propulsion rate.HE staining was used to detect physiological and pathological conditions of the gastric antrum tis-sue of rats.Western blot was used to detect the protein expression levels of p-AMPK/AMPK,p-ULK1/ULK1,FUNDC1,and LC3II/lC3I.ATP content in the gastric antrum tissue was detected using kits.Immunofluorescence staining was used to detect the content and co-localization level of LAMP2 and TOMM20 in the gastric antrum tissue of FD rats with a syndrome of liver depression and spleen deficiency.Results:No obvious organic changes were observed in the gastric an-trum tissue of any group.Compared with the blank control group,the model control group showed significant reductions in body weight,food intake,total movement distance,rearing frequency,gastric emptying rate,and small intestinal propul-sion rate(P<0.05).The protein expression levels of p-AMPK/AMPK,p-ULK1/ULK1,FUNDC1,and LC3II/lC3I were significantly increased(P<0.05).The fluorescence intensities of TOMM20 and LAMP2,as well as their co-localization coefficient,were also markedly increased(P<0.05),while the ATP content was significantly decreased(P<0.05).Compared with the model control group,the Shuwei Decoction groups and the Mosapride group showed significant im-provements in body weight,food intake,gastric emptying rate,small intestinal propulsion rate,total movement distance,and rearing frequency(P<0.05 or P<0.01).The protein expression levels of p-AMPK/AMPK,p-ULK1/ULK1,FUNDC1,and LC3II/lC3I were significantly decreased(P<0.05 or P<0.01).The fluorescence intensities of TOMM20 and LAMP2,as well as their co-localization coefficient,were significantly decreased(P<0.05 or P<0.01),while the ATP content was significantly increased(P<0.05 or P<0.01).Conclusion:The mechanism of action of Shuwei Decoc-tion on FD rats with a syndrome of liver depression and spleen deficiency is related to the regulation of mitochondrial au-tophagy via the AMPK/ULK1/FUNDC1 signaling pathway.
