丹龙定喘汤调节Th17/Treg细胞免疫平衡改善OVA诱导的支气管哮喘炎症OA
Improvement of OVA-Induced Asthmatic Airway Inflammation via Regulation of Th17/Treg Cell Immune Balance by Danlong Dingchuan Decoction
目的:探讨丹龙定喘汤对白蛋白(OVA)所致的支气管哮喘小鼠炎症的作用及机制.方法:60 只SPF级BALB/c雌性小鼠,随机分为正常对照组、模型对照组、地塞米松 0.63 mg/kg组、丹龙定喘汤 13、27、53 g/kg组,每组10 只.除正常对照组外,其余小鼠于第1、14 d腹腔注射0.2 mL OVA溶液致敏,在第28 天,将 1%OVA生理盐水溶液超声雾化并以最大雾量吸入激发,1 次/d,30 min/次,连续 5 d.第 35 天开始雾化吸入 1%OVA溶液激发哮喘,雾化3 次/周,同时各药物组灌胃给予相应药物,1 次/d,连续6 w.末次给药1 h后,小鼠检测气道反应性;检测结束后取材,摘眼球取血,全血测定血常规,观察炎症细胞变化;检测血清白介素 10(IL-10)、白介素 17(IL-17)的含量;取肺泡灌洗液检测辅助性T细胞 17(Th17 细胞)、调节性T细胞(Treg细胞)的水平;HE染色和Masson染色观察右肺病理和胶原沉积.结果:与正常对照组相比,模型对照组小鼠经不同浓度的乙酰甲胆碱刺激后Penh值显著升高,全血中白细胞和粒细胞比例显著增加,支气管周围炎症浸润与胶原沉积明显,肺泡灌洗液血清中Th17 细胞水平显著增加,Treg细胞水平、血清 IL-10 含量明显降低(P<0.05 或P<0.01);与模型对照组相比,丹龙定喘汤Penh值、全血中白细胞和粒细胞比例显著降低,支气管周围炎症浸润与胶原沉积情况显著改善,肺泡灌洗液中Th17 细胞水平、血清IL-17 含量显著降低,Treg细胞水平、血清IL-10 的含量明显升高(P<0.05 或P<0.01).结论:丹龙定喘汤能够有效改善OVA所致的哮喘模型小鼠的气道炎症反应,其机制可能与调节Th17/Treg间细胞免疫平衡进而影响炎症反应相关.
Objective:To explore the effect of Danlong Dingchuan Decoction(丹龙定喘汤)on ovalbumin(OVA)-induced asthmatic airway inflammation in mice and its underlying mechanism.Methods:Sixty SPF BALB/c female mice were randomly divided into six groups:normal group,model group,dexamethasone group(0.63 mg/kg),and groups receiving Danlong Dingchuan Decoction at low,medium,and high doses(13 g/kg,27 g/kg,and 53 g/kg,respectively),with 10 mice in each group.Except for the normal group,all other groups were sensitized by intraperitoneal injection of 0.2 mL OVA solution on days 1 and 14.On day 28,1%OVA saline solution was administered via ultrasonic nebulization at the maximum output once a day for 30 min over five consecutive days.From day 35 onward,mice were administered with 1%OVA solution via nebulization three times per week to stimulate asthma.Simultaneously,each treatment group received corresponding drugs by gavage once a day for six weeks.One hour after the last administration,the airway hyperrespons-iveness of mice was tested.After detection,the eyeballs were removed to collect blood.A complete blood count(CBC)was performed on whole blood to observe the changes in inflammatory cells.The serum levels of interleukin-10(IL-10)and interleukin-17(IL-17)were detected.Bronchoalveolar lavage fluid(BALF)was collected to detect the levels of Th17 and Treg cells.The right upper lung lobe was fixed with paraformaldehyde,embedded,and sectioned for hematoxy-lin-eosin(HE)staining and Masson's trichrome staining.Results:Compared with the normal group,the model group showed significantly increased Penh values after stimulation with different concentrations of methacholine,significantly el-evated proportion of white blood cells and granulocytes in whole blood,notable peribronchial inflammatory infiltration and collagen deposition,markedly increased proportion of Th17 cells in BALF and serum level of IL-17,and significantly de-creased proportion of Treg cells and serum level of IL-10(P<0.05 or P<0.01).Compared with the model group,all groups treated with Danlong Dingchuan Decoction showed significantly decreased Penh values,markedly reduced propor-tions of white blood cells and granulocytes in whole blood,notably alleviated peribronchial inflammatory infiltration and collagen deposition,markedly decreased Th17 cell proportion in BALF and serum IL-17 levels,and significantly increased Treg cell proportion and serum IL-10 level(P<0.05 or P<0.01).Conclusion:Danlong Dingchuan Decoction can effec-tively improve airway inflammation in OVA-induced asthma model mice,possibly by regulating the immune balance be-tween Th17 and Treg cells,thereby affecting the inflammatory response.
