线粒体基因组(Mitochondrial genome,mtDNA)对紫花苜蓿的遗传改良至关重要.本研究采用改良CTAB法从紫花苜蓿(品系为青大1号)叶中提取线粒体DNA并测序,结果表明青大1号紫花苜蓿mtDNA全长290 285 bp,包含35个蛋白质编码基因、15个tRNA和3个rRNA基因;鉴定出56个简单重复序列和48个长片段重复序列;多数蛋白编码基因受纯化选择(Ka/Ks<1),仅rps1和nad9基因(Ka/Ks>1)呈适应性进化;IR边界分析显示,其基因组较大豆等豆科植物更紧凑,SSR以四核苷酸重复为主;进化分析显示,其与蒺藜苜蓿亲缘关系最近;密码子偏好性分析表明,其偏好A/U结尾的密码子,中性绘图和ENC-plot显示自然选择对其偏好性影响较大.

Alfalfa(Medicago sativa L.)is an important leguminous forage grass,and characterization of its mitochondrial genome(mtDNA)is essential for understanding its evolutionary biology and supporting genetic improvement.In this study,the complete mtDNA sequence of the strain'Qingda No.1'was assembled and analyzed.The mtDNA is 290 285 bp in length and contains 35 protein-coding genes,15 transfer RNA(tRNA)genes,and three ribosomal RNA(rRNA)genes.A total of 56 simple sequence repeats(SSRs)and 48 long repeat sequences were identified.Analysis of synonymous and nonsynonymous substitution rates showed that most protein-coding genes were under purifying selection,with Ka/Ks ratio below 1,whereas rps1 and nad9 exhibited Ka/Ks ratio greater than 1,suggesting possible positive selec-tion.Inverted repeat(IR)boundary analysis indicated that the alfalfa mtDNA was more compact compared with other legumes such as soybean(Glycine max),and tetranucleotide repeats represented the predomi-nant SSR type.Phylogenetic analysis confirmed a close evolutionary relationship between M.sativa and Medicago truncatula.Codon usage bias analysis revealed a preference for codons ending with A or U,and neutrality and ENC-plot analyses indicated that natural selection played a major role in shaping codon usage bias.
